SVCT2-GLUT1-mediated ascorbic acid transport pathway in rat dental pulp and its effects during wound healing

Naoto Ohkura et al.

Summary: Ascorbic acid (vitamin C) plays a crucial role in promoting dental pulp wound healing by regulating cell differentiation and anti-inflammatory response. This study provides new insights into the mechanism of action of ascorbic acid and may contribute to the development of novel therapeutic targets for dental pulpal disease treatment.

Network analysis of large-scale ImmGen and Tabula Muris datasets highlights metabolic diversity of tissue mononuclear phagocytes

Anastasiia Gainullina et al.

Summary: This study focuses on the diversity and metabolic variability of mononuclear phagocyte (MNP) subpopulations. Through metabolic network analysis of large-scale transcriptional datasets, it was found that cholesterol synthesis is particularly active in migratory dendritic cells, while glutathione synthesis is essential for cysteinyl leukotriene production by peritoneal and lung macrophages.

Taurine enhances the antitumor efficacy of PD-1 antibody by boosting CD8+ T cell function

Yu Ping et al.

Summary: This study found that high serum taurine levels were associated with good response to PD-1 blockade antibody therapy in lung cancer patients. CD8(+) T cells cultured with exogenous taurine exhibited enhanced proliferation and secretion of cytotoxic cytokines. High SLC6A6 expression was correlated with an effector T cell signature in tumor patients. Taurine enhanced CD8(+) T cell proliferation and function through PLCγ1-mediated calcium and MAPK signaling. Combining taurine with immune checkpoint blockade antibody suppressed tumor growth and improved CD8(+) T cell function and proliferation.

Neuronal loss of NCLX-dependent mitochondrial calcium efflux mediates age-associated cognitive decline

Pooja Jadiya et al.

Summary: Mitochondrial calcium overload contributes to neurodegenerative disease development and progression. Loss of the mitochondrial sodium/calcium exchanger (NCLX) promotes calcium overload, metabolic derangement, redox stress, and cognitive decline in Alzheimer's disease models. Neuronal deletion of NCLX induces an Alzheimer's disease-like pathology, including memory decline, amyloid deposition, tau pathology, synaptic remodeling, and cell death.

SLC38A2 and glutamine signalling in cDC1s dictate anti-tumour immunity

Chuansheng Guo et al.

Summary: Cancer cells evade T cell-mediated killing through unknown mechanisms. Dendritic cells, especially type-1 conventional DCs, play a crucial role in T cell priming and therapeutic efficacy against tumors. The function of dendritic cells is orchestrated by pattern recognition receptors, but other signals involved are not fully defined. Nutrients have been identified as emerging mediators of adaptive immunity, but their impact on dendritic cell function and communication between innate and adaptive immune cells is still unclear.

Identification of Three Cuproptosis-specific Expressed Genes as Diagnostic Biomarkers and Therapeutic Targets for Atherosclerosis

Yong -tong Chen et al.

Summary: This study investigated the clinical implication of cuproptosis-related genes (CRGs) in atherosclerosis. Functional annotation and validation of selected genes were performed, and a signature of CRGs for diagnostic validation was constructed. The results suggest that the cuproptosis-related gene signature could serve as a potential diagnostic biomarker for atherosclerosis and may offer novel insights into the treatment of cardiovascular diseases.

Amino Acid Solute Carrier Transporters in Inflammation and Autoimmunity

Linlin Sheng et al.

Summary: This review summarizes the role of SLC amino acid transporters in inflammation and autoimmunity. The importance of Glutamate transporters SLC1A1, SLC1A2, and SLC1A3 in central nervous system disorders and the essential role of SLC7A1 and SLC7A2 in T cells and innate immunity are discussed. The link between SLC amino acid transporters and inflammation may provide potential therapeutic targets for related diseases.

Lactate Transporter SLC16A3 (MCT4) as an Onco-Immunological Biomarker Associating Tumor Microenvironment and Immune Responses in Lung Cancer

Qingsong Tao et al.

Summary: SLC16A3 is significantly upregulated in lung adenocarcinoma and squamous carcinoma, and its high expression is associated with poor prognosis. It plays a role in regulating immune cell infiltration and immunosuppression, and it may contribute to immune evasion through the HIE-1a-IL8 axis in the immune microenvironment.

Amino acid availability acts as a metabolic rheostat to determine the magnitude of ILC2 responses

Suzanne H. Hodge et al.

Summary: ILC2 are tissue-resident lymphocytes that respond rapidly to damage and infection at mucosal barrier sites. They are sensitive to cues from the diet and invading pathogens, and also to perturbations in nutrient availability. Amino acid availability has been identified as a nutritional cue in regulating ILC2 responses.

SLC7A8 is a key amino acids supplier for the metabolic programs that sustain homeostasis and activation of type 2 innate lymphoid cells

Santosh K. Panda et al.

Summary: ILC2 cells selectively express the Slc7a8 gene, which plays a crucial role in maintaining cellular fitness and activation. Lack of Slc7a8 leads to reduced amino acid availability, affecting energy metabolism and signaling pathway activation.
