Childhood Acute Lymphoblastic Leukemia: Results of the Randomized Acute Lymphoblastic Leukemia Intercontinental-Berlin-Frankfurt-MUnster 2009 Trial

Myriam Campbell et al.

Summary: The International BFM study group conducted a study on pediatric ALL, evaluating MRD using FCM and the impact of early intensification and MTX dose on survival. The results showed that MRD was successfully assessed using FCM, and a dose of 2g/m2 MTX was effective in preventing relapse in non-HR pcB-ALL. Augmented IB showed no advantages over the standard IB.

Prognostic value and multifaceted roles of tetraspanin CD9 in cancer

Robert Ondrussek et al.

Summary: CD9 is a crucial regulator of cell adhesion in the immune system, playing important physiological roles in hematopoiesis, blood coagulation, and infections. It is involved in leukocytes' transendothelial migration, which may be hijacked by cancer cells during invasion and metastasis. CD9 affects cancer progression and therapy resistance through its presence on cell surface and exosome membranes. The expression of CD9 is generally associated with good patient outcomes, but there are exceptions in breast, ovarian, melanoma, pancreatic, and esophageal cancer, possibly due to different antibodies or cancer heterogeneity. Studies on tetraspanin CD9's role in tumor suppression or promotion are inconclusive, requiring further mechanistic experiments.

Summary: Acute lymphoblastic leukemia (ALL) is the most common childhood cancer and has shown significant improvement in survival rates through risk-adapted chemotherapy, prevention measures, and molecular analysis. However, challenges remain for older patients and relapsed/refractory cases.

Prognostic factors for CNS control in children with acute lymphoblastic leukemia treated without cranial irradiation

Jingyan Tang et al.

Summary: An analysis of 7640 pediatric patients treated under the Chinese Children's Cancer Group ALL-2015 protocol identified male sex, BCR-ABL1 fusion in B-ALL, and high leukocyte count in T-ALL as risk factors for isolated CNS relapse. Additionally, the use of total intravenous anesthesia during intrathecal therapy and flow cytometry examination of diagnostic cerebrospinal fluid in B-ALL patients were associated with lower relapse rates.

Interaction between CD9 and PI3K-p85 activates the PI3K/AKT signaling pathway in B-lineage acute lymphoblastic leukemia

Yi-Fen Shi et al.

Summary: CD9 knockdown inhibits cell proliferation, migration, invasion, and promotes apoptosis, chemosensitivity in B-lineage acute lymphoblastic leukemia cells. CD9 activates the PI3K/AKT signaling pathway through direct interaction with PI3K-p85 in B-ALL cells.
