An influenza H1 hemagglutinin stem-only immunogen elicits a broadly cross-reactive B cell response in humans

Sarah F. Andrews et al.

Summary: Current seasonal influenza vaccines primarily target the constantly evolving hemagglutinin (HA) head region, resulting in limited cross-protection against other influenza strains or subtypes. In order to improve the immune response and offer broad protection, researchers developed a stabilized H1 stem immunogen displayed on a ferritin nanoparticle (H1ssF). A phase 1 clinical trial showed that vaccination with H1ssF induced a strong plasmablast response and sustained production of cross-reactive memory B cells targeting conserved epitopes on the HA stem. This demonstrates the potential for an H1 HA immunogen to elicit a robust and broadly neutralizing B cell response.

A chimeric hemagglutinin-based universal influenza virus vaccine approach induces broad and long-lasting immunity in a randomized, placebo-controlled phase I trial

Raffael Nachbagauer et al.

Summary: New influenza virus vaccines based on chimeric hemagglutinin have been found to induce a broad, strong, durable and functional immune response targeting the conserved stalk of the hemagglutinin protein. This suggests that these chimeric hemagglutinins have the potential to be developed as universal vaccines that protect broadly against influenza viruses.

Binding affinity landscapes constrain the evolution of broadly neutralizing anti-influenza antibodies

Angela M. Phillips et al.

Summary: Research has shown that two naturally isolated influenza broadly neutralizing antibodies (CR9114 and CR6261) exhibit different patterns of affinity, with CR9114 displaying significant constraints in affinity for different antigens and showing higher affinity in specific combinations, while CR6261 is less restricted in breadth to more similar antigens.
