The results revealed that Obacunone showed satisfactory results for density functional theory, molecular docking, and MD simulation study.

Original article Computer-assisted discovery of safe and effective DprE1/aaRSs Inhibitors against TB utilizing Drug Repurposing approach

Summary: In this study, a drug repurposing approach was employed to discover potential DprE1 inhibitors by conducting structure-based virtual screening of the FDA and world-approved drugs database. Through molecular docking, MMGBSA energy estimation, and ADMET prediction, three hit molecules with good binding interactions were identified. Molecular dynamics simulation revealed one of the hit molecules to be stable and possessing a known safety profile, suggesting its potential as a candidate for further optimization and development of new DprE1 inhibitors.

Summary: The study investigates the effects of electron-releasing and electron-withdrawing groups at para positions on the molecular structure of synthesized compounds in selected solvents and in the gas phase. Results show that intramolecular interactions between substituents and the electron donor-acceptor centers of the thiazole ring influence the relative stability of the compounds. Charge-transfer possibilities within the molecules are revealed through dipole moment values and energy gaps between occupied and unoccupied molecular orbitals.
