Streamlined alpha-amylase assays for wheat preharvest sprouting and late maturity alpha-amylase detection

Summary: Late maturity alpha-amylase (LMA) and preharvest sprouting (PHS) result in increased alpha-amylase in wheat grain. This study developed a high throughput 96-well method for small wheat grain samples using the Phadebas alpha-amylase enzyme assay, compared to FN and the Megazyme Alpha-Amylase SD Assay Kit. Low-cost small-scale milling methods were also evaluated. The Phadebas assay showed high reproducibility and strong correlation to the SD assay and FN method, offering higher sensitivity and lower expense.

The FN test indirectly detects the presence of alpha-amylase, which is responsible for starch degradation. Low FN/high alpha-amylase in wheat grains results in poor end-product quality and significant losses for various stakeholders. The FN method has limitations such as sampling variability, high cost, labor intensiveness, destructive nature, and inability to differentiate between different causes of low FN. Seeking faster, cheaper, and more accurate alternatives can improve breeding and avoid inadvertent mixing of high- and low-FN grain, thus preserving the value of wheat grain.
