Rational Design of N-Methylcarbamoylguanidinyl Derivatives as Highly Potent Dual-Target Chitin Hydrolase Inhibitors for Retarding Growth of Pest Insects

Summary: In this study, a series of compounds with N-methylcarbamoylguanidinyl and N-methoxycarbonylguanidinyl were designed and synthesized as dual-target inhibitors of Of ChtI and Of Hex1. The most potent dual-target inhibitor, compound 10d, showed IC50 values of 27.1 and 249.1 nM against Of ChtI and Of Hex1, respectively. Furthermore, insecticidal activity studies demonstrated significant effects of compounds 10a-c, 10k, and 10l on the growth and development of Plutella xylostella. This work provides a promising method for the development of novel chitin hydrolase inhibitors as potential pest control and management agents.

Metabolomic and Transcriptome Analysis of the Inhibitory Effects of Bacillus subtilis Strain Z-14 against Fusarium oxysporum Causing Vascular Wilt Diseases in Cucumber

Summary: Controlling cucumber Fusarium wilt caused by Fusarium oxysporum f. sp. cucumerinum (FOC) with the Bacillus subtilis strain Z-14 showed significant antagonistic activity, reaching a control effect of 88.46% in pot experiments. Microscopic observations revealed that strain Z-14 induced expansion and breakage of FOC hyphae, leading to uneven cell wall thickness and degraded organelle structure. Metabolome and transcriptome analysis demonstrated that strain Z-14 inhibited FOC infection by suppressing cell wall and cell membrane synthesis, energy metabolism, amino acid synthesis of FOC mycelium, as well as ROS clearance and CWDEs secretion, thereby affecting MAPK signal transduction and transport function inhibition.

Synthesis and Fungicide Activity on Asperisporium caricae of Glycerol Derivatives Bearing 1,2,3-Triazole Fragments

Summary: This study describes the preparation and fungicidal activity evaluation of 12 glycerol derivatives bearing 1,2,3-triazole fragments. The results showed that the glycerol derivatives significantly inhibited conidial germination of Asperisporium caricae, the etiological agent of papaya black spot. Compound 4c exhibited the most active inhibitory effect and improved the severity of black spots on papaya fruits.

Exploration of the dynamic interplay between lipids and membrane proteins by hydrostatic pressure

Double-activation of mitochondrial permeability transition pore opening via calcium overload and reactive oxygen species for cancer therapy

Summary: In this study, a mitochondrial pathway was established to inhibit tumor growth through the double activation of the mitochondrial permeability transition pore (MPTP) channel. Fe2+ doped covalent organic frameworks (COF) were synthesized and used as templates to grow a CaCO3 shell, resulting in the formation of O-2-FeCOF@CaCO3 nanocomposite. After modification with folic acid (FA), the nanocomposite (OFCCF) could target breast cancer cells and achieve synergistic treatment through photodynamic therapy (PDT) and calcium overload. The multifunctional nanosystem with tumor microenvironment (TME) response abilities provided a novel strategy for clinical cancer therapy.

Structure-Activity Studies of N-Heterocyclic Benzoyl Arylamine Derivatives Led to a Highly Fungicidal Candidate against Gaeumannomyces graminis var. tritici and Four Fusarium Wheat Pathogens

The results showed that the position and structure of the N-heterocyclic group and the substituted group on the benzamide scaffold significantly influenced the activities. Compound 18f exhibited excellent activities against multiple wheat root pathogens, making it a potential novel fungicide candidate for controlling wheat root diseases.

Summary: An increase in permeability of the mitochondrial inner membrane, referred to as mitochondrial permeability transition (PT), plays a central role in cell death and tissue damage. The permeability transition pore (PTP), which is induced by calcium and cyclosporin A (CSA), is believed to be responsible for PT, although the molecular details are not fully understood. This study used holographic and fluorescent microscopy to investigate the contribution of ATP synthase and adenine nucleotide translocator (ANT) to PTP. The findings suggest that both ATP synthase and ANT are required for high-conductance PTP but not depolarization.

Activity of Fengycin and Iturin A Isolated From Bacillus subtilis Z-14 on Gaeumannomyces graminis Var. tritici and Soil Microbial Diversity

Summary: This study demonstrated that fengycin and iturin A purified from Bacillus subtilis Z-14 showed strong antifungal effects against Gaeumannomyces graminis var. tritici (Ggt), the pathogen causing wheat take-all disease. In biocontrol tests, both fengycin and iturin A effectively controlled the disease, with fengycin treatment significantly affecting soil bacterial diversity after 7 days. Additionally, the two compounds led to changes in the relative abundance of other fungal genera in the soil.
