Nanofibrillar peptide hydrogels for self-delivery of lonidamine and synergistic photodynamic therapy

Summary: This study designs and synthesizes a self-assembling peptide molecule (LND-K) that targets both integrin receptors and mitochondria of cancer cells, leading to enhanced ablation of mitochondrial function in melanoma cells. By co-assembling LND-K with an anionic photosensitizer TPPS4, a nanofibrillar hydrogel system is developed for slow release and prolonged retention of TPPS4, achieving synergistic killing of cancer cells in vitro and in vivo.

A Polymeric Extracellular Matrix Nanoremodeler for Activatable Cancer Photo-Immunotherapy

Summary: Cancer immunotherapy has potential in the clinic, but the extracellular matrix (ECM) in the tumor microenvironment presents a barrier. A semiconducting polymer-based ECM nanoremodeler (SPNcb) is reported to combine photodynamic antitumor activity with the inhibition of collagen-crosslinking enzymes for cancer photo-immunotherapy. SPNcb blocks collagen crosslinking and decreases ECM stiffness, improving the tumor infiltration of cytotoxic T cells and inhibiting tumor growth and metastasis.

Enzyme-triggered deep tumor penetration of a dual-drug nanomedicine enables an enhanced cancer combination therapy

Summary: Emerging nanomedicines can eliminate cancer cells by promoting excessive intracellular reactive oxygen species (ROS) generation. However, tumor heterogeneity and poor penetration of nanomedicines often result in varying levels of ROS production, where low levels of ROS can actually promote tumor cell growth, reducing the effectiveness of these nanomedicines. In this study, a novel amphiphilic and block polymer-dendron conjugate-derived nanomedicine (GFLG-DP/Lap NPs) was developed, incorporating a photosensitizer (Ppa) for ROS therapy and Lapatinib (Lap) for molecular targeted therapy. By combining ROS therapy with Lap, the synergistic effect effectively kills cancer cells, even in deep tumor regions.
