Intestinal bacteria and colorectal cancer: etiology and treatment

Michael W. Dougherty et al.

Summary: The development of colorectal cancer is influenced by the presence of bacterial communities in the gastrointestinal tract. These microorganisms obtain essential nutrients from indigestible compounds and play a role in molecular signaling pathways that are necessary for normal tissue and immune function. Several bacterial species, such as Fusobacterium nucleatum, Enterotoxigenic Bacteroides fragilis, and pks(+) E. coli, have been identified to potentially increase the risk of colorectal cancer. Recent research has expanded on this topic to include host mutational status, microbial heterogeneity within tumors, transient infections, and the cumulative effects of multiple carcinogenic bacteria. This review provides an updated overview of how host-bacteria interactions influence colorectal cancer development, potential applications in diagnosis or prevention, and the impact of the gut microbiome on colorectal cancer treatment efficacy.

Deficiency of SR-B1 reduced the tumor load of colitis-induced or APCmin/+-induced colorectal cancer

Qijun Chen et al.

Summary: Scavenger receptor class B type 1 (SR-B1) knockdown reduces tumor load in colitis-induced or APCmin/+-induced colorectal cancer (CRC). SR-B1 knockdown improves immune microenvironment and gut microbiota by regulating levels of tumor-associated macrophages, mononuclear myeloid-derived suppressor cells, granulocytic myeloid-derived suppressor cells, and programmed cell death-ligand 1. SR-B1 knockdown triggers anti-CRC effect of anti-PD 1 in CRC.

CircEZH2/miR-133b/IGF2BP2 aggravates colorectal cancer progression via enhancing the stability of m(6)A-modified CREB1 mRNA

Bing Yao et al.

Summary: This study identified a novel circular RNA called circEZH2 in colorectal cancer tissues, which is closely related to the clinical characteristics and prognosis of colorectal cancer patients. circEZH2 promotes the proliferation and migration of colorectal cancer cells, and regulates the expression of IGF2BP2 by interacting with m(6)A reader IGF2BP2 and acting as a sponge for miR-133b. Additionally, circEZH2/IGF2BP2 enhances the stability of CREB1 mRNA, thereby aggravating colorectal cancer progression.

The involvement of gut microbiota in the anti-tumor effect of carnosic acid via IL-17 suppression in colorectal cancer

Siyu Li et al.

Summary: In this study, the researchers investigated the role of carnosic acid (CA) in the development of colorectal cancer (CRC). They found that CA regulated the gut microbiota and serum metabolites, and inhibited CRC development through the NF-kappa B/STAT3 signaling pathway. CA also suppressed inflammation, contributing to its anti-CRC properties.

Targeting cathepsin B by cycloastragenol enhances antitumor immunity of CD8 T cells via inhibiting MHC-I degradation

Guoliang Deng et al.

Summary: This study reveals that Cycloastragenol (CAG) has anti-tumor effects by promoting tumor cell-surface antigen presentation and enhancing the killing function of CD8(+) T cells. Mechanistically, CAG binds to cathepsin B, inhibiting the lysosomal degradation of MHC-I and promoting its aggregation on the cell membrane, which enhances the presentation of tumor antigens. Furthermore, the combination of CAG and PD-1 antibodies enhances the tumor-killing ability of CD8(+) T cells.

New Baitouweng decoction combined with fecal microbiota transplantation alleviates DSS-induced colitis in rats by regulating gut microbiota metabolic homeostasis and the STAT3/NF-κB signaling pathway

Xin Gu et al.

Summary: This study aimed to investigate the synergistic effect and potential mechanism of New Baitouweng Decoction (NBD) combined with fecal microbiota transplantation (FMT) in rats with DSS-induced ulcerative colitis (UC). The results showed that the combination of NBD and FMT significantly improved the disease condition, tight junction protein expression, and macrophage marker expression in UC rats. Furthermore, NBD combined with FMT increased short-chain fatty acid concentrations and inhibited the activation of related proteins. Microbiota sequencing results indicated that the diversity and abundance of the gut microbiota were improved after treatment with NBD and FMT.

Summary: Tumorigenesis is influenced by interactions between cancer cells and the tumor microenvironment. Tumor-associated macrophages play a crucial role in tumor development, with the potential to both promote antitumor responses and exhibit immunosuppressive protumor phenotypes. Targeting TAMs has become a strategy in cancer therapy, with a focus on reprogramming TAMs to support antitumor immune responses.

Summary: The study found that co-administration of total polysaccharides from Astragalus membranaceus and Codonopsis pilosula could improve clinical symptoms, restore immune balance, and alleviate colonic mucosal damage in colitis mice. The unique efficacy of co-administration depended on the activation of the aryl hydrocarbon receptor (AhR) and upregulation of isovaleric acid and butyrate at specific doses.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
