Recent progress in nanocarrier-based drug delivery systems for antitumour metastasis

Summary: Tumor metastasis is a major contributor to poor prognosis and lower survival rates in cancer patients. While surgery alone is often insufficient to treat distant metastatic tumors, chemotherapy can improve healing rates and patient prognosis. However, the effectiveness of chemotherapy drugs is limited by their low solubility, lack of tumor targeting, instability and toxicity, as well as multidrug resistance. Immunotherapy has shown promise but is associated with adverse reactions and acquired resistance. Fortunately, the use of nanocarriers to deliver chemotherapeutic drugs can reduce systemic reactions, inhibit metastasis, and improve therapeutic outcomes.

Summary: This study demonstrates a self-assembled strategy for simultaneous delivery of the therapeutic drug salinomycin (SAL) and siRNA to cancer cells using chitosan nanoparticles. The siRNA@C-SAL showed superior cytotoxicity against gastric carcinoma cells without significant adverse effects in healthy mice. Thus, this improved drug delivery system with siRNA and hydrophobic drugs provides a potential strategy for the treatment of gastric cancer.

NIR phosphorescent cyclometalated platinum (II) complexes with CAIX targeted and nuclear penetration as potent anticancer theragnostic agents

Summary: This study developed a series of cyclometalated platinum (II) complexes with near-infrared phosphorescent emission targeting carbonic anhydrase IX (CAIX). These complexes showed high binding affinity and effective inhibition on CAIX in vitro. The complexes demonstrated significantly enhanced therapeutic effect on multiple cancer cell lines compared to cisplatin and had higher selectivity towards cancer cells.

Effects of p-sulfonatocalixarene and p-sulfonatocalixarene/sulfobetaine surfactant complex on the activities of bromelain and polyphenol oxidase

Summary: Calixarene, a third-generation supra-molecular compound, has potential applications in clinical disease diagnosis and intervention treatment. In this study, the binding of sulfonatocalix[n]arene (SC[n]) with bromelain (BM) and polyphenol oxidase (PPO) was investigated in the absence and presence of sulfobetaine surfactant (SDDAB). SC[n] entered the docking pocket of proteins in a 1:1 molar ratio, while the proteins did not enter the cavity of SC[n]. SC[n] formed a stronger bond with BM than with PPO and resulted in a greater inhibition of BM activity. SC[n] and SDDAB formed complexes, and the proteins interacted with the SC[n]/SDDAB complex instead of disrupting the binding between SC[n] and SDDAB. The protein activities could be controlled by adjusting the molar ratio of SC[n] to the surfactant.

Self-assembled dehydropeptide nanocarrier as a delivery system for antitumor drug temozolomide

Summary: In this study, stable molecular conformation and intermolecular forces were demonstrated to be essential for peptide self-assembly. A novel dehydropeptide monomer was successfully prepared and self-assembled into a nanomaterial driven by hydrogen bonding. The resulting dehydropeptide nanomaterial exhibited excellent biocompatibility, resistance to enzymatic hydrolysis, and improved drug loading efficiency. The loaded antitumor drug showed enhanced entrapment efficiency, prolonged half-life, and increased antitumor activity against cancer cells, highlighting the potential of biocompatible nanocarriers in drug delivery systems.

Summary: Breast cancer stem cells (BCSCs) are a major contributor to the unfavorable outcomes seen in many breast cancer patients, as they are more prevalent in hypoxic environments and refractory to traditional chemotherapy. A small molecule construct named AzCDF has been developed to target BCSCs effectively in the hypoxic tumor environment, reducing tumor growth and migration. This is the first time a CSC-targeting small molecule has been shown to prevent tumorigenesis in an animal model.
