Perspective: The viscoelastic properties of biofilm infections and mechanical interactions with phagocytic immune cells

Marilyn Wells et al.

Summary: Biofilms, as viscoelastic materials, are a major public health problem and a cause of chronic bacterial infections due to their resistance to the immune system's clearance. However, the relationship between the mechanical properties of biofilms and their resistance to phagocytosis remains largely unexplored. This article provides an overview of biofilm infections and their interactions with the immune system, biofilm mechanics, and highlights the importance of studying this field to discover targets for therapeutics that can enhance the immune system's efficacy.

Bacterial Biofilm Formation on Biomaterials and Approaches to Its Treatment and Prevention

Panxin Li et al.

Summary: Bacterial biofilms can cause widespread infections and have strong resistance to antibiotics. This article summarizes the formation processes, resistance mechanisms, and main characteristics of clinically persistent infections caused by biofilms. Additionally, two main methods for preventing and treating biofilm-related infections on biomaterials are introduced: antibacterial coatings and surface modification of biomaterials. The advantages of each strategy in terms of antibacterial effect, biocompatibility, limitations, and application prospects are analyzed, providing ideas and research directions for the development of novel biofilm infection strategies related to therapeutic materials.

Mucoid Pseudomonas aeruginosa Can Produce Calcium-Gelled Biofilms Independent of the Matrix Components Psl and CdrA

Holly M. Jacobs et al.

Summary: This study reveals the diverse nature of biofilm formation and structure in Pseudomonas aeruginosa. Mucoid P. aeruginosa develops biofilms using a novel program that is dependent on calcium and does not rely on the canonical biofilm EPS components. These mucoid biofilms impede the penetration of antibiotics and display a unique arrangement of individually suspended cells. The findings highlight the compositional and structural variety of P. aeruginosa biofilm aggregates.

Microrheology of Pseudomonas aeruginosa biofilms grown in wound beds

Minhaz Ur Rahman et al.

Summary: A new technique was used to measure the viscoelasticity of in vivo Pseudomonas aeruginosa biofilms. The results show that the biofilms grown in vivo are relatively more elastic and exhibit similar compliance compared to those grown in vitro. Additionally, the contributions of exopolysaccharides to biofilm viscoelasticity were different for the biofilms grown in vitro and in vivo. The incorporation of host material, particularly collagen, into the matrix of the biofilm when it is grown in vivo may be the dominant contributor to biofilm viscoelasticity.

Adaptation and Evolution of Pathogens in the Cystic Fibrosis Lung

Paul J. Planet

Summary: In contrast to acute respiratory infections, persistent bacterial infections in cystic fibrosis (CF) allow bacteria to evolve and adapt over time. This adaptation is reflected in mutations that accumulate in the genomes of the infecting bacteria. These mutations can lead to phenotypic differences such as antibiotic resistance or changes in the mucoid phenotype of Pseudomonas aeruginosa. Many of these adaptations occur repeatedly in different patients and even across different bacterial species.

Opportunistic Pathogens in Cystic Fibrosis: Epidemiology and Pathogenesis of Lung Infection

Ana C. Blanchard et al.

Summary: Cystic fibrosis (CF) is a common genetic disease in Caucasians, leading to respiratory failure caused by chronic infections. The respiratory infections in CF patients are caused by opportunistic pathogens such as Pseudomonas aeruginosa, Burkholderia spp., Achromobacter spp., Stenotrophomonas maltophilia, anaerobes, nontuberculous mycobacteria, and fungi. Culture-independent methods have revealed the polymicrobial nature and dynamics of microbial communities in CF lung infections. The unique environment of the CF airway facilitates infections by opportunistic pathogens. Infectious disease physicians should be aware of the differences in epidemiology and disease role of these pathogens in CF compared to other medical conditions.

Neutrophils: Many Ways to Die

Erandi Perez-Figueroa et al.

Summary: Neutrophils play a crucial role in innate immune response by executing various effector functions. When activated, they undergo cell death processes to ensure the proper elimination of pathogens and damaged cells, contributing to timely resolution of inflammation. Neutrophils' cell death mechanisms include apoptosis, pyroptosis, necrosis, and others, ultimately maintaining homeostasis in the organism.

Cation complexation by mucoid Pseudomonas aeruginosa extracellular polysaccharide

Oliver J. Hills et al.

Summary: The study creates two large mucoid P. aeruginosa EPS models based on beta-D-mannuronate and beta-D-mannuronate-alpha-L-guluronate systems, encompassing thermodynamically stable acetylation configurations unique to mucoid P. aeruginosa. Using first principles calculations, the study identifies stable coordination environments of cations in CF lungs and quantifies their thermodynamic stability. The models demonstrate the weaker cross-linking capability of Na+ and Mg2+ ions compared to Ca2+ ions, with a preference for cation binding within M-G blocks due to smaller torsional rearrangements required to reveal stable binding sites. The geometry of chelation site influences the stability of resulting complexes more than electrostatic interactions, providing nuanced chemical insight into previous experimental observations.
