Hypermethylated GRIA4, a potential biomarker for an early non-invasive detection of metastasis of clinically known colorectal cancer

Eva Lukacova et al.

Summary: A sensitive droplet digital PCR (ddPCR) assay was developed to evaluate the diagnostic potential of GRIA4 hypermethylation in colorectal cancer (CRC) patients. The study found that GRIA4 methylation was significantly increased in tumor tissues and showed promising diagnostic ability. Furthermore, high levels of GRIA4 hypermethylation were also observed in the blood samples of patients with metastatic CRC, suggesting its potential for non-invasive early detection.

Role of solute carrier transporters SLC25A17 and SLC27A6 in acquired resistance to enzalutamide in castration-resistant prostate cancer

Prem P. Kushwaha et al.

Summary: SLC25A17 and SLC27A6 play a crucial role in the development of enzalutamide resistance through metabolic reprogramming. Inhibition of these proteins suppresses cell proliferation and migration, reduces fatty acid synthesis and lactic acid levels, and may serve as therapeutic targets to overcome drug resistance.

Colorectal cancer promoter methylation alteration affects the expression of glutamate ionotropic receptor AMPA type subunit 4 alternative isoforms potentially relevant in colon tissue

Ana Florencia Vega-Benedetti et al.

Summary: This study evaluated the expression of two main isoforms of GRIA4 in CRC, finding downregulation of both variants in tumors, while the short isoform showed higher expression at the protein level. This discrepancy may be explained by miRNA specifically targeting the canonical isoform, suggesting a functional role of the short isoform in intestinal cell biology.

A comprehensive analysis of the diagnostic and prognostic value associated with the SLC7A family members in breast cancer

Liping Yan et al.

Summary: This study investigated the expression and prognostic value of SLC7 family genes in breast cancer. The results revealed that high SLC7A5 expression was associated with poorer survival time, while SLC7A4 was associated with good progression-free interval and disease-specific survival. SLC7A3 expression was positively associated with overall survival and distant metastasis-free survival. SLC7A family genes were strongly correlated with T helper cells and natural killer cells, suggesting their role in modulating immune infiltration in breast cancer.

Summary: NMDAR signaling induces chemoresistance to TMZ via upregulation of MGMT expression in GBM cells. MEM enhances the cytotoxic effect of TMZ and reduces MGMT expression by inhibiting NMDAR, increasing DNA alkylation by TMZ in MGMT-positive cells.

Multi-omic machine learning predictor of breast cancer therapy response

Stephen-John Sammut et al.

Summary: The response to therapy in breast cancers is influenced by the pre-treated tumour ecosystem, and predictive models integrating multi-omics features through machine learning can be used to predict treatment outcomes. The degree of residual disease following therapy is monotonically associated with pre-therapy features of the tumour.

Breast cancer resistance to chemotherapy: When should we suspect it and how can we prevent it?

Prihantono et al.

Summary: Chemotherapy is a crucial treatment for breast cancer, but chemoresistance poses a challenge to its effectiveness. Factors such as drug inactivation, changes in drug targets, and cancer stem cells contribute to chemoresistance. Clinical and pathological evaluations are used to assess chemotherapy effectiveness, with methods including physical examination and tumor residue analysis. Additional approaches for predicting and preventing chemoresistance involve molecular processes and metabolic activity evaluation, as well as using the right combination of chemotherapy methods and drugs.

Chemoresistance and Metastasis in Breast Cancer Molecular Mechanisms and Novel Clinical Strategies

Jun Cao et al.

Summary: Breast cancer is the most common malignant tumor in females worldwide, and chemotherapy is the standard treatment. However, chemoresistance is a common issue with metastatic breast cancer, and the mechanisms underlying chemoresistance and metastasis have not been fully explored. Possible molecular mechanisms of chemoresistance include efflux transporters, signaling pathways, non-coding RNAs, and cancer stem cells. Novel clinical strategies like drug carriers, immunotherapy, and autophagy regulation are being researched to overcome this challenge.
