Immune checkpoint inhibitors: Key trials and an emerging role in breast cancer

Nicola Gaynor et al.

Summary: This article reviews the use of immune checkpoint inhibitors as immunomodulatory agents for relieving immune suppression caused by tumors. These inhibitors have shown impressive results in various solid cancers, especially melanoma and non-small cell lung cancer. They have potential benefits for most cancer types and numerous clinical studies are being conducted to explore their efficacy. The article provides an overview of key clinical trials involving immune checkpoint inhibitors targeting PD-1, PD-L1, and CTLA-4, and discusses emerging therapies targeting these checkpoints as well as the status of immune checkpoint inhibitors in breast cancer.

CAR-T Cell Therapy for Breast Cancer: From Basic Research to Clinical Application

Yu-Huan Yang et al.

Summary: Breast cancer became the most commonly diagnosed cancer in 2020, and CAR-T cell therapy, an immunotherapy, has shown limited efficacy in treating breast cancer. This review discusses the development of CAR-T cell therapy in breast cancer, including tumor-associated antigens, ongoing clinical trials, obstacles to its effectiveness, and potential strategies for improvement.

The balance between breast cancer and the immune system: Challenges for prognosis and clinical benefit from immunotherapies

Constantin N. Baxevanis et al.

Summary: Cancer evolution is a complex process influenced by genetic factors and external stimuli that trigger signaling pathways between tumor cells and the immune system. The immune system plays a dual role in cancer progression, conveying protective antitumor immunity while also facilitating tumor development by shaping tumor immunogenicity. Understanding the interaction between immune elements and tumor cells within the tumor microenvironment is crucial for prognosis and developing immunotherapy strategies for various cancers.

The relationship between hypoxia-inducible factor 1α (HIF-1α) and patient survival in breast cancer: Systematic review and meta-analysis

Suad A. K. Shamis et al.

Summary: The analysis confirmed a significant association between high expression of HIF-1 alpha and poorer disease-free survival (DFS) and overall survival (OS) in breast cancer patients. Various factors such as survival analysis, study region, antibodies used, scoring methods, and threshold methods were found to impact the expression of HIF-1 alpha.

21-Gene Assay to Inform Chemotherapy Benefit in Node-Positive Breast Cancer

Kevin Kalinsky et al.

Summary: The study showed that among premenopausal women with hormone-receptor-positive, HER2-negative breast cancer and one to three positive lymph nodes with a recurrence score of 25 or lower, the addition of chemotherapy to endocrine therapy led to longer invasive disease-free survival and distant relapse-free survival compared to endocrine therapy alone. However, postmenopausal women with similar characteristics did not benefit from adjuvant chemotherapy in terms of invasive disease-free survival.

Cancer-Associated Adipocytes in Breast Cancer: Causes and Consequences

Ilona Rybinska et al.

Summary: Breast cancer progression is heavily influenced by the interaction between tumor cells and cancer-associated adipocytes (CAAs), and understanding the molecular pathways related to adipocyte differentiation is crucial for devising new therapeutic interventions. Recent studies have highlighted the mechanisms through which CAAs impact breast cancer progression, including adipokine regulation, metabolic reprogramming, extracellular matrix remodeling, and immune cell modulation.

Macrophage Polarization States in the Tumor Microenvironment

Ava J. Boutilier et al.

Summary: The M1/M2 macrophage paradigm is crucial in tumor progression, where M1 macrophages have anti-tumor effects and M2 macrophages contribute to tumor development through various mechanisms. The tumor microenvironment plays a role in influencing macrophage polarization and activation, which is essential for understanding the pro-tumor effects of tumor-associated macrophages.

Adjuvant Olaparib for Patients with BRCA1- or BRCA2-Mutated Breast Cancer

Andrew N. J. Tutt et al.

Summary: In patients with high-risk, HER2-negative early breast cancer and germline BRCA1 or BRCA2 pathogenic or likely pathogenic variants, adjuvant olaparib after completion of local treatment and neoadjuvant or adjuvant chemotherapy was associated with significantly longer survival free of invasive or distant disease than placebo. Olaparib had limited effects on global patient-reported quality of life.
