ZNF460 mediates epithelial-mesenchymal transition to promote gastric cancer progression by transactivating APOC1 expression

Liangliang An et al.

Summary: This study revealed the significant up-regulation of ZNF460 in gastric cancer, and its expression correlated with tumor grade, lymph node metastasis, and H. pylori infection. Functionally, diminished ZNF460 expression inhibited gastric cancer cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) in vitro, and suppressed tumor growth in vivo. Mechanistically, ZNF460 combined with apolipoprotein C1 (APOC1) promoter to facilitate APOC1 transcription and accelerated EMT, thereby promoting gastric cancer progression.

Levels of pretreatment serum lipids predict responses to PD-1 inhibitor treatment in advanced intrahepatic cholangiocarcinoma

Zhenyun Yang et al.

Summary: This study aimed to clarify the prognostic value of serum lipids and establish an effective nomogram for predicting overall survival in iCCA patients receiving anti-PD1 therapy. The results showed that serum lipid levels were associated with patient prognosis, with APOA1 and TG being significant independent predictors for overall survival in multivariate analysis. By constructing a predictive nomogram, the survival rate of iCCA patients can be more accurately predicted.

BAL Proteomic Signature of Lung Adenocarcinoma in IPF Patients and Its Transposition in Serum Samples for Less Invasive Diagnostic Procedures

Summary: Idiopathic pulmonary fibrosis (IPF) is a chronic and irreversible fibrosing interstitial pneumonia of unknown cause. IPF increases the risk of lung cancer (LC), and the two diseases share similar risk factors and pathogenic pathways. A proteomic study on bronchoalveolar lavage fluid (BALF) samples identified dysregulated proteins related to inflammation, immune response, lipid metabolism, and cell adhesion in LC associated with IPF (LC-IPF) patients. These potential biomarkers could potentially improve the management of LC-IPF.

Elevated expression of ISY1, APOA-1, SYNE1, MTG1, and MMP10 at HCC initiation: HCC specific protein network involving interactions of key regulators of lipid metabolism, EGFR signaling, MAPK, and splicing pathways

Laila H. Faraj Shaglouf et al.

Summary: This study aimed to identify molecular regulators of hepatocellular carcinoma (HCC) initiation and progression using a combination of protein and gene expression analyses. The researchers induced HCC in male Wistar rats and characterized differentially expressed proteins in early stage HCC tissues. They also analyzed the mRNA expression of these proteins and their interacting partners, as well as the molecular mechanisms associated with signaling pathways involved in HCC progression. The study identified an upregulated protein called ISY1, which is associated with HCC initiation, progression, and tumor stages, and validated its interacting partners. The dysregulation of mRNA and protein expression of these regulators may play a critical role in HCC initiation and progression.

Apolipoprotein E induces pathogenic senescent-like myeloid cells in prostate cancer

Nicolo Bancaro et al.

Summary: Tumor cells promote the recruitment of immunosuppressive neutrophils, a subset of myeloid cells driving immune suppression, tumor proliferation, and treatment resistance. We have identified a subset of neutrophils that have upregulated expression of cellular senescence markers and persist in the tumor microenvironment. These senescent-like neutrophils express TREM2 and are more immunosuppressive and tumor-promoting than canonical immunosuppressive neutrophils.

Cholesterol efflux pathways hinder KRAS-driven lung tumor progenitor cell expansion

Emma Guilbaud et al.

Summary: Cholesterol efflux pathways play a role in tumor biology and can be targeted as a metabolic therapy for lung cancer. Disruption of these pathways in epithelial progenitor cells promotes tumor growth and creates a pro-tolerogenic tumor microenvironment. Increasing HDL levels protects against tumor development by inhibiting the positive feedback loop between growth factor signaling and cholesterol efflux pathways. Cholesterol removal therapy using cyclodextrin reduces tumor burden by suppressing the proliferation and expansion of tumor progenitor cells.

Summary: Apolipoprotein E (ApoE) is a multifunctional protein that plays a role in lipid transport, inflammation regulation, and immune function. Its genetic polymorphisms are associated with the risk of degenerative diseases and its overexpression has been found in various solid tumors. ApoE can regulate macrophage polarization, tumor immune microenvironment, inflammation, and tumor progression, invasion, and metastasis. However, further research is needed to fully understand the functions of ApoE and its relationship with diseases.

Involvement of clusterin expression in the refractory response of pancreatic cancer cells to a MEK inhibitor

Kohei Amada et al.

Summary: Constitutive activation of the MAPK signaling pathway is necessary for PDAC tumorigenesis, but targeting this pathway with inhibitor treatment has not yielded positive clinical results. By studying the response of PDAC cells to the MAPK inhibitor PD0325901, it was found that overexpression of clusterin (CLU) played a role in the development of resistance to the inhibitor. PD0325901 and CLU downregulation together showed synergistic or additive effects in reducing PDAC cell proliferation. Overexpression of CLU was also observed in surgically resected PDAC tissues. These findings suggest a potential therapeutic strategy for PDAC patients using inhibitors targeting the MAPK signaling pathway and CLU.

Metabolic syndrome and thyroid Cancer: risk, prognosis, and mechanism

Ling-Rui Li et al.

Summary: The increasing incidence of thyroid cancer (TC) cannot be fully explained by overdiagnosis. Metabolic syndrome (Met S) is highly prevalent due to the modern lifestyle, which can lead to the development of tumors. This review discusses the relationship between Met S and TC, including risk, prognosis, and potential biological mechanisms. The association between Met S and TC risk and aggressiveness has been observed, with gender differences in most studies. Abnormal metabolism and the influence of hormones play key roles in tumorigenesis, and markers of metabolic disorders are expected to aid in diagnosis and prognosis. Targeting specific signaling pathways may provide new treatment options for TC.

TRIM15 promotes the invasion and metastasis of pancreatic cancer cells by mediating APOA1 ubiquitination and degradation

Yan Sun et al.

Summary: Pancreatic ductal adenocarcinomas (PDACs) are often diagnosed at an advanced stage, with metastasis posing a challenge to patient survival. The TRIM15 gene is implicated in cancer development, particularly in PDAC metastasis regulation. High TRIM15 expression in PDAC tissues is associated with poor prognosis and its interaction with APOA1 promotes lipid anabolism, potentially aiding in PDAC metastasis. Targeting the TRIM15-APOA1-LDLR axis may be a strategy to inhibit PDAC metastasis by blocking triglyceride synthesis.
