Cartilage extracellular matrix-derived matrikines in osteoarthritis

Anna E. Rapp et al.

Summary: Osteoarthritis (OA) is a common musculoskeletal disease characterized by degradation of cartilage extracellular matrix (ECM). The release of intact/full-length proteins and proteolytic fragments during this degradation process can induce different downstream responses, leading to various biological consequences. Collagen type II and aggrecan are the major components of cartilage ECM, but numerous minor components have also been identified, some of which have been shown to play a role in OA. There is currently no effective therapy for OA, and understanding the matrikine landscape in joint tissues may help identify biomarkers and potential drug targets for disease modification.

Advanced glycation end products in musculoskeletal system and disorders

Akinobu Suzuki et al.

Summary: The global aging population and the increase in diabetes cases have led to a high prevalence of musculoskeletal disorders in elderly and diabetic patients, which are associated with advanced glycation end products (AGEs). AGEs accumulate in tissues and contribute to various aging and diabetes-related conditions such as osteoporosis, osteoarthritis, and neuropathy. Understanding the mechanisms of AGEs can potentially lead to the development of novel prevention and therapy methods for these disorders, improving patients' quality of life.

Summary: The present study focuses on the preparation of a polymer-based material with incorporated ceramics and the monitoring of changes in properties after natural degradation in vitro. The developed material is a mixture of polylactic acid and polyhydroxybutyrate, with 10% hydroxyapatite and 10% tricalcium phosphate incorporated. The results show that the water absorption, weight loss, and mechanical properties of the material change with increasing degradation time.

Mechanosensitive Control of Articular Cartilage and Subchondral Bone Homeostasis in Mice Requires Osteocytic Transforming Growth Factor β Signaling

Karsyn N. Bailey et al.

Summary: The study demonstrates the involvement of osteocytic TGF beta signaling in OA, affecting joint homeostasis, SBP thickness, and sclerostin expression. The findings suggest a sexually dimorphic role of osteocytic TGF beta signaling in joint homeostasis and mechanosensation at the SBP. Osteocytic TGF beta signaling could be a potential therapeutic target for OA.
