Discovery of Diverse CRISPR-Cas Systems and Expansion of the Genome Engineering Toolbox

Summary: CRISPR systems in bacteria and archaea mediate adaptive immunity through diverse effector mechanisms. They have been repurposed for versatile applications in therapeutics and diagnostics thanks to their facilereprogramming with RNA guides. RNA-guided CRISPR-Cas targeting and interference are mediated by effectors that are either components of multisubunit complexes in class 1 systems or multidomain single-effector proteins in class 2. The rich diversity of CRISPR enzymes has led to rapid maturation of the genome editing toolbox, with capabilities ranging from gene knockout to RNA editing. Combined with rational design and engineering, the natural diversity of CRISPR and related bacterial RNA-guided systems provides a vast resource for expanding the repertoire of tools for molecular biology and biotechnology.

Efficient Genome and Base Editing in Human Cells Using ThermoCas9

Summary: This study discovers a small, naturally accurate, and thermostable type II-C Cas9 ortholog called ThermoCas9 from Geobacillus thermodenitrificans. It demonstrates that ThermoCas9 is active in human cells and can be used as an efficient genome editing tool, especially for gene disruption. The researchers also develop a ThermoCas9-mediated base editor, called ThermoBE4, which exhibits a three times larger window of activity compared to the traditional SpyCas9 base editor (BE4), providing an alternative platform to expand the targeting scope of both genome and base editing in human cells.

Programmable endonuclease combined with isothermal polymerase amplification to selectively enrich for rare mutant allele fractions

Researchers have recently used programmable endonucleases to enrich mutant alleles and improve detection. This strategy has the potential to reduce the cost and time of cancer screening and genotyping, and enable targeted therapies in resource-limited settings.

Natural Nucleoside Modifications in Guide RNAs Can Modulate the Activity of the CRISPR-Cas9 System In Vitro

Summary: In this study, we show that incorporating N-6-methyladenosine, 5-methylcytidine, and pseudouridine modifications into guide RNAs enhances the efficiency and specificity of the CRISPR-Cas9 system. We found that the complexes of modified guide RNAs with Cas9 protein promote cleavage of target duplexes and plasmid substrates, while reducing immunostimulatory and cytotoxic effects of sgRNAs.

DNA topology regulates PAM-Cas9 interaction and DNA unwinding to enable near-PAMless cleavage by thermophilic Cas9

Summary: A Cas9 protein (Alicyclobacillus tengchongensis) from a thermophilic bacterium has been found to regulate PAM recognition through DNA topology. Structural analysis reveals that AtCas9 has a unique loop motif that interacts with the DNA major groove to modulate PAM recognition. Experimental results demonstrate the near-PAMless editing capability of AtCas9 in E. coli and its broad PAM preference for efficient editing in mammalian cells.

Unraveling the Genomic Potential of the Thermophilic Bacterium Anoxybacillus flavithermus from an Antarctic Geothermal Environment

Summary: Thermophilic organisms have been extensively studied in various fields, such as industrial biotechnology and exploration of life limits. This study successfully developed a thermostable CRISPR-Cas9 system for genome editing of thermophiles, demonstrating targetable nuclease activity at temperatures up to 65 degrees C. The results of this research may facilitate the development of genome editing tools for other extreme thermophiles, as the vector has been made available for further research and use.
