Molecular Genetic Analysis of Ukrainian Families with Congenital Cataracts

Xiaodong Jiao et al.

Summary: This study aims to identify pathogenic variants causing autosomal dominant congenital cataracts in five Ukrainian families. Whole-exome sequencing revealed heterozygous disease-causing variants in HSF4, CRYGA, GJA3, CRYGC, and PAX6. These variants were confirmed to cosegregate with cataracts using Sanger sequencing. Overall, four novel mutations and one previously reported mutation were found to be responsible for autosomal dominant congenital cataracts.

Whole Exome Sequencing in Coloboma/Microphthalmia: Identification of Novel and Recurrent Variants in Seven Genes

Patricia Haug et al.

Summary: Coloboma and microphthalmia (C/M) are related congenital eye malformations that exhibit significant genetic heterogeneity, as revealed by a study utilizing high throughput DNA sequencing technologies. The study identified novel gene variants and potentially disease-causing genes in a portion of C/M patients, including three de novo mutations. This highlights the diagnostic power of whole-exome sequencing for genetic screening in patients and families with C/M.

Cataract-causing mutations L45P and Y46D impair the thermal stability of γC-crystallin

Chenxi Fu et al.

Summary: This study investigated the thermal stability of gamma C-crystallin mutants and found that L45P and Y46D mutations impaired the stability. However, alpha A-crystallin could rescue the thermal stability of these mutants to some extent.

beta B2 W151R mutant is prone to degradation, aggregation and exposes the hydrophobic side chains in the fourth Greek Key motif

Jingjie Xu et al.

Summary: Congenital cataract is a major cause of blindness in children globally. A novel beta B2 mutation W151R was found to be prone to aggregation due to low solubility and poor structural stability, but the negative effect of the mutation was reversed by administration of lanosterol.

Crystalline gene mutations in Turkish children with congenital cataracts

Mine Karahan et al.

Summary: The study aimed to detect crystallin gene mutations in Turkish children with congenital cataracts. A total of 56 patients were included, with a male-to-female ratio of 2.1:1 and an age range from 2 months to 5 years. Gene mutations were detected in 7% of the patients, with one novel mutation and three known mutations identified.
