VIP interneuron impairment promotes in vivo circuit dysfunction and autism-related behaviors in Dravet syndrome

Kevin M. Goff et al.

Summary: DS is a severe neurodevelopmental disorder caused by loss-of-function variants in SCN1A. In this study, the impaired function of VIP-INs in DS mice was observed, and optogenetic activation of VIP-INs restored neuronal activity. These findings suggest that the dysfunction of VIP-INs may underlie the cognitive and behavioral comorbidities in DS.

DSCAM Deficiency Leads to Premature Spine Maturation and Autism-like Behaviors

Peng Chen et al.

Summary: Mutations in cell adhesion molecules (CAMs) can lead to abnormal synapse formation and maturation, possibly contributing to autism spectrum disorders (ASDs). This study discovered that the DSCAM gene, a high-risk gene for autism, plays a role in preventing premature spine maturation and excessive glutamatergic transmission. Deficiency of DSCAM led to accelerated dendritic spine maturation and autism-like behaviors in mice.

Spine impairment in mice high-expressing neuregulin 1 due to LIMK1 activation

Peng Chen et al.

Summary: The high-level expression of NRG1 leads to reduced dendritic spine density and decreased frequency of miniature excitatory postsynaptic currents. This is accompanied by activation of LIMK1 and increased cofilin phosphorylation in postsynaptic densities.
