Non-Structural Protein 3 of Duck Tembusu Virus Induces Autophagy via the ERK and PI3K-AKT-mTOR Signaling Pathways

Summary: Despite the economic losses caused by duck Tembusu virus (DTMUV) to the poultry industry, the specific relationships between DTMUV and cellular autophagy are largely unknown. This study investigated the interactions between autophagy and DTMUV and found that DTMUV increased autophagy, which in turn facilitated viral replication. Non-structural protein 3 (NS3) was identified as the main protein inducing autophagy, and this induction was mediated by the ERK2, PI3K/AKT, and mTOR signaling pathways. NS3-induced autophagy promoted DTMUV replication.

SARS-CoV-2 Nsp14 mediates the effects of viral infection on the host cell transcriptome

Summary: Viral infection involves a complex set of events orchestrated by multiple viral proteins. In this study, the researchers focused on the functions of Nsp14, a protein involved in viral RNA replication. They found that the expression of Nsp14 caused significant changes in the transcriptome and splicing of genes, as well as an increase in circRNAs. These effects were independent of Nsp14's exonuclease activity and required the N7-guanine-methyltransferase domain of the protein. Activation of the NFkB pathway and increased expression of CXCL8 were observed early upon Nsp14 expression. The researchers identified IMPDH2 as a key mediator of these effects, suggesting its involvement in guanine nucleotides biosynthesis. Overall, this study reveals a previously unknown role for Nsp14 with implications for therapy.

GFAT1-linked TAB1 glutamylation sustains p38 MAPK activation and promotes lung cancer cell survival under glucose starvation

Summary: This study reveals an unidentified mechanism of p38 MAPK signaling regulation by metabolic enzyme during nutrient stress. The interaction between GFAT1 and TAB1 promotes the formation of the GFAT1-TAB1-p38 signaling pathway, which in turn promotes autophagy and tumor cell survival under glucose deficiency. Clinical analysis shows that GFAT1 and TAB1 S438 phosphorylation levels correlate with the prognosis of lung adenocarcinoma patients.
