Towards Bioinspired Meniscus-Regenerative Scaffolds: Engineering a Novel 3D Bioprinted Patient-Specific Construct Reinforced by Biomimetically Aligned Nanofibers

Summary: This study presents a new biofabrication strategy for developing biomimetic meniscus scaffolds. The use of bioprinted cell-laden hydrogel reinforced by aligned nanofiber sheets improved the structural integrity, shape fidelity, and mechanical properties of the scaffolds. Importantly, the introduction of nanofiber mats did not negatively affect cell viability. This bioengineering approach offers a promising strategy for tissue engineering of the knee meniscus.

Touchspinning: Mechanically drawing polyacrylonitrile nanofibers

Summary: Electrospinning is a common method for producing polymer nanofibers, but it has challenges. Mechanical drawing of nanofibers is easier, but scaling up is limited. The new Touchspinner device can continuously cold-draw nanofibers, with efficiency comparable to electrospinning but fewer drawbacks. Controllable parameters have been found to affect the size of the nanofibers, with optimal parameters resulting in highly aligned nanofibers.

Understanding Fibroblast Behavior in 3D Biomaterials

Summary: Fibroblasts exhibit diverse behaviors in different culture systems and their response to biomimetic materials is influenced by a complex interplay of biological features, material properties, and microarchitectural features. Understanding the influence of biomaterials on fibroblast behavior is challenging due to the heterogeneous nature of fibroblasts and their tissue-specific phenotypic traits. Despite these challenges, investigating fibroblast behavior in 3D culture using a range of materials can provide insights into potential therapeutic opportunities for diseases such as cancer and fibrosis.

Xeno-free bioengineered human skeletal muscle tissue using human platelet lysate-based hydrogels

Summary: Bioengineered human skeletal muscle tissues have been developed as in vitro systems for disease modeling. In this study, a-PL-based nanocomposite hydrogels were used as scaffolds to encapsulate human muscle satellite stem cells. The modulation of nanocrystal content facilitated the formation of highly aligned muscle fibers.

Coral-Derived Collagen Fibers for Engineering Aligned Tissues

Summary: This study introduces a new biomaterial system based on centimeter-long collagen fibers extracted from Sarcophyton soft corals, wrapped around frames to create aligned fiber arrays. Cytotoxicity tests demonstrated that the collagen arrays were nontoxic to fibroblast cells.

Potential of laponite incorporated oxidized alginate-gelatin (ADA-GEL) composite hydrogels for extrusion-based 3D printing

Summary: A new composite hydrogel combining oxidized alginate-gelatin (ADA-GEL) hydrogel and laponite nanoclay was successfully developed and characterized in this study. The addition of 0.5% laponite nanoplatelets improved the printability of ADA-GEL hydrogels. Different needle types (cylindrical and conical) were compared in filament fusion test, showing enhanced printing accuracy with a cylindrical tip. A glass flip test determined a processing window of 1-2 hours after composite ink preparation, confirming laponite/ADA-GEL hydrogel composites as promising inks for 3D bioprinting.

Oscillatory rheology of carboxymethyl cellulose gels: Influence of concentration and pH

Soft Elastic Fibrous Scaffolds for Muscle Tissue Engineering by Touch Spinning

Summary: This study presents a new approach for fabricating highly aligned soft elastic fibrous scaffolds, demonstrating their potential for muscle tissue engineering applications. By synthesizing polyester-polyurethane soft copolymers with different molecular weights and chain extenders, materials with varied physicochemical and mechanical properties were obtained. The selected materials offer advantages such as biocompatibility, biodegradability, suitable mechanical properties, high recovery ratio, and characteristics mimicking the extracellular matrix of muscle tissue.
