However, their poor bioavailability and lack of controlled-release mechanisms have been addressed by scaffolding them with biomaterials to create Exosomes-loaded scaffolds (ELS). These biomaterial-based scaffolds can be designed and functionalized to improve the bioavailability, biocompatibility, and controlled release of Exosomes.

Curcumae Rhizoma Exosomes-like nanoparticles loaded Astragalus components improve the absorption and enhance anti-tumor effect

Summary: This study aimed to improve the absorption of Astragalus components (AC) using plant exosomes and demonstrate the synergistic mechanism of AC-loaded Curcumae Rhizoma Exosomes-like nanoparticles (AC-CELNs). CELNs containing active ingredients from Curcumae Rhizoma were extracted and loaded with Formononetin, Astragaloside IV, and Calycosin from Astragalus. AC-CELNs showed slow drug release and improved stability against gastric and intestinal fluids. They also exhibited superior uptake and transport capacity in cell models, and the endocytosis pathways involved clathrin- and caveolin-dependent mechanisms. AC-CELNs significantly decreased cell survival in a time- and concentration-dependent manner. Additionally, pharmacokinetic studies in rats showed that CELNs promoted the in vivo absorption of AC. These findings suggest that CELNs are promising carriers for enhancing the absorption and anti-tumor activity of AC.

Inhibitory mechanism of phenolic compounds in rapeseed oil on α-amylase and α-glucosidase: Spectroscopy, molecular docking, and molecular dynamic simulation

Summary: The important phenolic compounds from rapeseed oil were found to effectively inhibit a-amylase and a-glucosidase. The inhibition mechanism was non-competitive and involved hydrogen bonds, hydrophobic force, or 7L-7L interaction. These compounds-induced conformation alterations of the enzymes were confirmed by FT-IR and molecular dynamics.

In this study, Res-encapsulated tetrasulfide-containing organosilica nanoparticles (DSMSNs) were developed as an oral delivery carrier, and the targeted delivery of Res to colon epithelial cells and macrophages was achieved through the use of chondroitin sulfate (CS) and a glutathione (GSH) response-mediated disassembly.

Natural exosome-like nanovesicles from edible tea flowers suppress metastatic breast cancer via ROS generation and microbiota modulation

Summary: This study developed a new therapeutic strategy using natural nanovehicles derived from tea flowers to trigger apoptosis of breast tumors and inhibit their lung metastasis. The nanovehicles had desirable particle sizes, exosome-like morphology, and contained polyphenols, flavonoids, functional proteins, and lipids. The experiments showed that the nanovehicles exhibited strong cytotoxicity against cancer cells by generating reactive oxygen species (ROS). In addition, in mice investigations, the nanovehicles were found to accumulate in breast tumors and lung metastatic sites after injection or oral administration, resulting in inhibition of breast cancer growth and metastasis, as well as modulation of gut microbiota.
