Serum levels of the novel adipokine isthmin-1 are associated with obesity in pubertal boys

Summary: This study aimed to evaluate the association between serum levels of the novel insulin-like adipokine isthmin-1 (ISM1) and obesity-related phenotypes in Spanish children, as well as investigate the molecular alterations behind the alteration of ISM1 levels in children with obesity. The results showed that boys with obesity had higher serum ISM1 levels compared to normal weight and overweight boys. ISM1 levels were positively associated with BMI Z-score and fat mass in boys, and negatively associated with myeloperoxidase (MPO). Although no associations were found in girls, fat mass was positively associated with ISM1 levels in all children. DNA methylation levels of two CpG sites in ISM1 were associated with serum levels of ISM1 in children. In conclusion, ISM1 is associated with obesity in boys during puberty and may serve as a biomarker of obesity in children.

Summary: This study reveals the importance of alveolar macrophages (AMs) in maintaining lung homeostasis and their role in chronic obstructive pulmonary disease (COPD). The absence of IST1 protein leads to increased AM numbers and altered functions, resulting in lung inflammation and COPD-like symptoms. The study also identifies csGRP78high AMs as the primary targets of IST1-mediated apoptosis. Intratracheal delivery of recombinant IST1 shows potential as a therapeutic strategy for COPD by targeting csGRP78.
