Clauson-Kaas pyrrole synthesis using diverse catalysts: a transition from conventional to greener approach

Dileep Kumar Singh et al.

Summary: This article presents the progress in the synthesis of N-substituted pyrrole derivatives using environmentally friendly methods. It summarizes the reaction using various acid and transition metal catalysts, involving the reaction of pyrrole with different aliphatic/aromatic primary amines and sulfonyl primary amines in the presence of 2,5-dimethoxytetrahydrofuran. The review evaluates the use of diverse conventional and greener catalysts in the modified Clauson-Kaas reaction for the synthesis of N-substituted pyrrole derivatives.

Catalytic activity in vitro of the human protein kinase ASK1 mutants: Experimental and molecular simulation study

Yayu Xie et al.

Summary: Kinases have been successfully targeted for drug discovery, but drug resistance is a concern. Assessing kinase mutations is essential for developing next-generation inhibitors. In this study, two mutants of ASK1 were constructed and their catalytic activity was investigated. Experimental results showed decreased binding affinity for the mutants compared to the wild type, and molecular dynamics simulations revealed the loss of hydrogen bond interaction as a possible cause for the decreased affinity.

A modification-centric assessment tool for the performance of chemoproteomic probes

Ji-Xiang He et al.

Summary: Chemoproteomics is an important technology for exploring fundamental biology and discovering new small-molecule-based therapies. pChem is a computational tool that can assess the performance of chemoproteomic probes by recognizing isotopically coded probe-derived modifications, calculating masses accurately, and localizing sites precisely. The tool exports reports that score the profiling efficiency, modification homogeneity, and proteome-wide residue selectivity of the tested probe.

Molecular docking and dynamics based approach for the identification of kinase inhibitors targeting PI3Kα against non-small cell lung cancer: a computational study

Debojyoti Halder et al.

Summary: This study used computational drug discovery tools to investigate non-small cell lung cancer (NSCLC) and aimed to identify potential protein kinase inhibitors through molecular docking and dynamics approach. The research discovered that molecules 6943 and 34100 showed better results compared to the PI3K inhibitor Copanlisib in docking assessment, ADMET analysis, and molecular dynamics simulation.

Protecting group-free method for synthesis of N-glycosyl carbamates and an assessment of the anomeric effect of nitrogen in the carbamate group

Yoshiyasu Ichikawa et al.

Summary: The first protecting group-free synthesis of N-glycosyl carbamates was achieved by reacting D-glucose with n-butyl carbamate in acidic aqueous media. The structures of the N-glucosyl carbamates were determined by comparison with authentic samples, and an anomeric pair of N-xylopyranosyl carbamates were prepared to assess the anomeric effect of nitrogen in the carbamate group.

Min-Ran Wang et al.

Summary: In this study, a new Paal-Knorr agent ONAyne was developed to investigate the reactivity and selectivity of gamma-dicarbonyl warheads in biological systems. Utilizing an unbiased open-search strategy, the lysine specificity of ONAyne on a proteome-wide scale was demonstrated, along with the characterization of various probe-derived modifications. Additionally, the method allowed for global, in situ, and site-specific profiling of targeted lysine residues of specific isomers of isoketals, revealing functional interactions in macrophages.
