Impact of lactic acid bacteria and their metabolites on the techno-functional properties and health benefits of fermented dairy products

Summary: The fermentation process of lactose to lactic acid in fermented dairy products leads to biochemical changes such as improved protein digestibility, release of fatty acids, and production of bioactive compounds. Various lactic acid bacteria are used to develop fermented dairy products with specific desirable attributes. These bacteria produce bioactive compounds and metabolites using homo-fermentative and heterofermentative pathways. The metabolites generated during fermentation vary with incubation and processing conditions and are analyzed and quantified using advanced instrumentation. The health benefits of fermented dairy products are mainly attributed to the biological roles of these metabolites, which also contribute to technological improvements desired by dairy manufacturers and consumers.

Beyond probiotics: a narrative review on an era of revolution

Summary: Whether intentionally or not, humans have been consuming probiotic microorganisms through fermented foods for many generations. Probiotics, such as lactic acid bacteria, are generally considered harmless and beneficial for human health. However, their therapeutic use is limited due to being living organisms. As a result, the focus has shifted to non-viable microbes made from probiotics, such as paraprobiotics, postbiotics, psychobiotics, nutribiotics, and gerobiotics, which can still provide health benefits and reveal therapeutic effects. This review summarizes the new terminology, traits, and applications of these microbes.

Bifidobacterium longum CCFM1206 Promotes the Biotransformation of Glucoraphanin to Sulforaphane That Contributes to Amelioration of Dextran-Sulfate-Sodium-Induced Colitis in Mice

Summary: This study screened probiotics with the ability to metabolize glucoraphanin and investigated the effects of a combination of B. longum CCFM1206 and BSE on colitis induced by DSS in mice. The results showed that the combination treatment could alleviate colitis symptoms, reduce inflammatory reactions and oxidative stress, and protect the intestinal barrier. Furthermore, the combination increased the production of sulforaphane compounds in the colon and activated genes related to the Nrf2 signaling pathway. These findings suggest that probiotics can enhance the bioactivity of dietary phytochemicals in vivo and have potential therapeutic implications for colitis treatment.

Dietary fiber and SCFAs in the regulation of mucosal immunity

Summary: Gut bacterial metabolites, specifically short-chain fatty acids (SCFAs), have significant effects on immune cells and gut health. Insufficient fiber intake hampers the production of SCFAs, leading to compromised immune functions and Western lifestyle diseases. There is a need to reconcile the role of fiber intake and the hygiene hypothesis in the development of these diseases. Mechanisms by which SCFAs influence immunity and gut homeostasis include activation of specific receptors, inhibition of histone deacetylases, and metabolic changes. Modulating SCFA levels through dietary interventions holds potential for prevention and treatment of diseases.

Bile acids and their receptors in regulation of gut health and diseases

Bile acids undergo microbial transformation by gut microbes and have profound effects on liver, intestinal tissues, and the gut microbiota. Farnesoid X receptor and G protein-coupled bile acid receptor 1, highly expressed in the intestine, have emerged as pivotal regulators in bile acid metabolism, innate immunity, and inflammatory responses. Manipulating bile acid metabolism and the expression of bile acid receptors may offer a promising strategy to regulate intestinal health and disease. This review aims to summarize the roles of bile acids and their receptors in the regulation of gut health and diseases.

Lactobacillus plantarum-derived indole-3-lactic acid ameliorates colorectal tumorigenesis via epigenetic regulation of CD8+ T cell immunity

Summary: Previous studies have shown that probiotics can ameliorate colorectal cancer in a mouse model, but the underlying mechanisms are still unclear. In this study, the researchers found that a specific probiotic strain and its metabolite could improve intestinal inflammation, tumor growth, and gut dysbiosis. Further investigation revealed that the metabolite enhanced the production of IL12a in dendritic cells, priming CD8+ T cell immunity against tumor growth, and also regulated cholesterol metabolism in CD8+ T cells. These findings provide new insights into the epigenetic regulation of probiotics-mediated anti-tumor immunity and suggest potential therapeutic strategies for colorectal cancer patients.

Summary: The aim of this study was to investigate the potential of Bifidobacterium infantis fermentation to alter the composition and physiological properties of barley juices. B. infantis JFM12 demonstrated a strong ability to decrease the total sugar content within 24 hours of fermentation. The volatile metabolite profiles were enriched as a result of B. infantis JFM12 fermentation, leading to changes in various compounds. Additionally, the antioxidant and lipase inhibitory activities of fermented barley juice were higher compared to unfermented barley juice.

Two doses of Lactobacillus induced different microbiota profiles and serum immune indices in pigs

Summary: We studied the impact of Lactobacillus reuteri ZLR003 supplementation on the microbiota and serum immune indices of growing pigs. The Lactobacillus doses had significant effects on the pigs' growth performance. L. reuteri ZLR003 was found in the gastrointestinal tract and showed increased species richness and Bacteroidetes abundance. Several bacterial abundances were significantly altered in the Lactobacillus-treated groups compared to controls. The Lactobacillus-treated pigs also exhibited changes in acid concentrations and serum immune markers. Our findings have implications for the use of Lactobacillus strains in pig production.

Ursodeoxycholic acid reduces antitumor immunosuppression by inducing CHIP-mediated TGF-β degradation

From Function to Metabolome: Metabolomic Analysis Reveals the Effect of Probiotic Fermentation on the Chemical Compositions and Biological Activities of Perilla frutescens Leaves

Summary: This study investigated the impact of probiotic fermentation on the active components and functions of Perilla frutescens leaves (PFL). The results showed that most of the active component contents and functional activities of PFL exhibited an increasing and then decreasing trend during fermentation. Different probiotics had distinguishable effects on PFL, and fermentation with ATCC 53013 for 1 day showed the highest enhancement effect. Metabolomic analysis revealed significant metabolite changes under the best fermentation condition, involving the generation of fatty acids and their conjugates, flavonoids. The study provides insights into the action mechanism of probiotic fermentation on the chemical composition and biological activity of PFL.

Modulation of the Gut Microbiota and Glycometabolism by a Probiotic to Alleviate Amyloid Accumulation and Cognitive Impairments in AD Rats

Summary: Regulating the gut microecology with probiotics is an effective strategy for the prevention and treatment of Alzheimer's disease (AD). This study demonstrates that a functional probiotic, Lactobacillus plantarum MA2, isolated from traditional Chinese Tibetan kefir grains, can improve cognitive deficits and anxiety-like behaviors in AD rats. It also attenuates neuronal degeneration and Aβ accumulation in the brain. Furthermore, the study shows that MA2 alleviates intestinal mucosal impairments and inhibits microglia activation and neuroinflammation through the TLR4/MYD88/NLRP3 signaling pathway. The findings suggest that MA2 reshapes the gut microbiota and modulates glycometabolism, and its exopolysaccharides (EPS) have inhibitory effects on Aβ aggregation and cytotoxicity induced by amyloid.

Isolation, characterization and anti-UVB irradiation activity of an extracellular polysaccharide produced by Lacticaseibacillus rhamnosus VHPriobi O17

Summary: This study aimed to evaluate the anti-UVB viability of exopolysaccharides (EPS) isolated from lactic acid bacteria (LAB). A LAB strain, Lacticaseibacillus rhamnosus VHPriobi O17, with high EPS production was selected. The purified EPS (OP-2) was identified as a novel Man-like polysaccharide with potential anti-UVB effects. It effectively alleviated cell apoptosis and reactive oxygen species production induced by UVB, inhibited the release of pro-inflammatory cytokines, and suppressed the phosphorylation cascade to prevent cell apoptosis. These findings suggest that EPS produced by L. rhamnosus VHPriobi O17 may be useful for human protection against UVB irradiation.

Pediococcus acidilactici FZU106 alleviates high-fat diet-induced lipid metabolism disorder in association with the modulation of intestinal microbiota in hyperlipidemic rats

Summary: This study investigated the effects of Pediococcus acidilactici FZU106, isolated from traditional brewing of Hongqu rice wine, on lipid metabolism and intestinal microbiota in high-fat diet-induced hyperlipidemic rats. The results showed that the intervention of P. acidilactici FZU106 effectively improved body weight, serum and liver parameters related to lipid metabolism and oxidative stress. It also demonstrated positive effects on the composition and metabolism of gut microbiota. Additionally, P. acidilactici FZU106 regulated the expression of liver genes involved in lipid metabolism and bile acid homeostasis. These findings suggest that P. acidilactici FZU106 has the potential to reduce disturbances in lipid metabolism by modulating both intestinal microflora and liver gene expression profiles.

Lactobacillus kefiranofaciens JKSP109 and Saccharomyces cerevisiae JKSP39 isolated from Tibetan kefir grain co-alleviated AOM/DSS induced inflammation and colorectal carcinogenesis

Summary: This study investigated the therapeutic effects of Lactobacillus kefiranofaciens JKSP109 (LK) and Saccharomyces cerevisiae JKSP39 (SC) on colon inflammation and colorectal carcinogenesis. The results showed that administration of LK, SC, and their combination improved body weight, decreased disease activity index, and reduced tumor multiplicity. These treatments also positively regulated gut microbiota, enhanced gut barrier function, reduced inflammation, and promoted apoptosis of tumor cells. Effects of exopolysaccharide from Lactobacillus rhamnosus on human gut microbiota in in vitro fermentation model
