Retinopathy of prematurity: A review of pathophysiology and signaling pathways

Summary: Retinopathy of prematurity (ROP) is a vasoproliferative disorder of the retina that leads to visual impairment and childhood blindness worldwide. The disease involves retinal microvascular degeneration, neovascularization, and potential retinal detachment. Oxidative and nitrosative stress, inflammatory processes, as well as nitric oxide synthase and arginase, play important roles in ROP progression. Mediators like vascular endothelial growth factor and succinate contribute to neovascularization, while extracellular matrix and vasorepulsive molecules like semaphorin 3A are involved in the pathogenesis. This article highlights the signaling pathways and molecular mediators of ROP, aiming to develop preventive and therapeutic strategies.

One-stop assembly of adherent 3D retinal organoids from hiPSCs based on 3D-printed derived PDMS microwell platform

Summary: We have successfully differentiated three-dimensional retinal organoids (ROs) with growth and development similar to the human retina from human induced pluripotent stem cells (hiPSCs) using a novel polydimethylsiloxane (PDMS) microwell platform. Our results showed that the ROs could uniformly adhere and differentiate into mature cells, maintaining homogeneity over a long period of time. We also established a xeno-free culture workflow by replacing fetal bovine serum with human platelet lysate, facilitating clinical application.

Anti-angiogenic properties of rapamycin on human retinal pericytes in an in vitro model of neovascular AMD via inhibition of the mTOR pathway

Summary: This study aimed to investigate the effect of inhibiting the mammalian target of rapamycin (mTOR) on the angiogenic cellular pathways in primary human retinal pericytes (HRP) using an in vitro model. The results showed that rapamycin efficiently inhibited proliferation and migration of the HRP cells, and disrupted the formation of endothelial cell tube structures in the presence of pericytes.
