Cellular and molecular insights into incomplete immune recovery in HIV/AIDS patients

Liting Yan et al.

Summary: Highly active antiretroviral therapy (ART) can effectively inhibit HIV replication and restore immune function. However, a significant number of patients do not achieve satisfactory increases in CD4(+) T cell counts, leading to incomplete immune reconstitution or immunological nonresponse (INR). INR patients have an increased risk of clinical progression and higher mortality rates. The mechanisms underlying INR are still unclear, despite extensive research. This review aims to explore the cellular and molecular factors contributing to incomplete immune reconstitution.

Neuroinflammatory transcriptional programs induced in rhesus pre-frontal cortex white matter during acute SHIV infection

Chase E. Hawes et al.

Summary: Research highlights the immune surveillance of the rhesus CNS by innate and adaptive immune cells during homeostasis. Following SHIV infection, vulnerabilities of key cognitive and motor function brain regions to HIV-1 during the acute phase were demonstrated by viral entry and integration within multiple brain regions.

Tripterygium wilfordii: An inspiring resource for rheumatoid arthritis treatment

Yanqiong Zhang et al.

Summary: Tripterygium wilfordii Hook F (TwHF) therapy is efficient for treating rheumatoid arthritis (RA) but can induce multi-organ toxicity. Guidelines have been developed to standardize TwHF-based therapy, yet crucial gaps in knowledge remain as potential barriers. More interdisciplinary attention is needed to enhance therapeutic effects while ensuring safety.

The role of CD38 in HIV infection

Liqi Lu et al.

Summary: CD38 is a widely expressed molecule that plays important roles in regulating cell differentiation and activation states, participating in various physiological and pathological processes. Studies have shown its association with HIV infection and its potential to impact disease progression through different mechanisms. Investigating CD38's role in HIV infection and related signaling pathways could offer new perspectives on potential treatments.

Increased homeostatic cytokines and stability of HIV-infected memory CD4 T-cells identify individuals with suboptimal CD4 T-cell recovery on-ART

Maria Pino et al.

Summary: Clinical outcomes are inferior in individuals with HIV experiencing suboptimal CD4 T-cell recovery during ART. The study found that immunologic suboptimal responders (ISR) had higher HIV-DNA levels in multiple CD4 T-cell subsets and higher expression of PD-1 and TIGIT exclusively on CM and TM CD4 T-cells pre-ART, indicating higher HIV reservoir maintenance. Additionally, ISR showed a lack of response to IL-7 and IL-15, potentially contributing to suboptimal CD4 T-cell recovery during ART.
