Cytoskeletal orchestration of glucose uptake in Sertoli cell to support efferocytosis of apoptotic germ cells

Di Wu et al.

Summary: This study reveals the correlation between beta II-spectrin and glucose metabolism as well as cytoskeletal structure in Sertoli cells. Deficiency of beta II-spectrin impairs glucose uptake and lactate production in Sertoli cells, leading to disrupted cytoskeleton and blood-testis barrier integrity, ultimately affecting efferocytosis. The findings suggest a potential link between beta II-spectrin deregulation and male infertility.

Deneddylation of PML/RARα reconstructs functional PML nuclear bodies via orchestrating phase separation to eradicate APL

Xuejing Shao et al.

Summary: Acute promyelocytic leukemia (APL) is driven by the oncoprotein PML/RAR alpha, which disrupts the construction of PML nuclear bodies (NBs). The disrupted NB assembly in APL is caused by neddylation-induced aberrant phase separation of PML/RAR alpha. Deneddylation restores the phase separation process, suppresses leukemogenesis, and can be targeted for APL eradication.

NFkB-signaling promotes glial reactivity and suppresses Muller glia-mediated neuron regeneration in the mammalian retina

Summary: Muller glia (MG) in mammalian retinas are incapable of regenerating neurons after damage, whereas the MG in lower vertebrates regenerate functional neurons. Identification of cell signaling pathways and gene regulatory networks that regulate MG-mediated regeneration is key to harnessing the regenerative potential of MG. In this study, NFkB signaling was found to play a crucial role in regulating immune cell accumulation and suppressing the neurogenic potential of MG after damage. Inhibition of NFkB enhanced the reprogramming of MG into neuron-like cells. Additionally, TGF beta 2 signaling and suppression of NFI and Id transcription factors were coordinated with NFkB signaling to regulate MG-mediated regeneration.

A subthreshold synaptic mechanism regulating BDNF expression and resting synaptic strength

Patricia M. Horvath et al.

Summary: Recent studies have shown that protein translation can be regulated by spontaneous excitatory neurotransmission, while the impact of spontaneous neurotransmitter release on gene transcription remains unclear. Blocking spontaneous inhibitory events increases the transcription of BDNF and Npas4 through altered synaptic calcium signaling, which can be prevented by antagonism of NMDA receptors or L-type voltage-gated calcium channels. Transcription can be bidirectionally altered by manipulating spontaneous inhibitory, but not excitatory, currents. Moreover, blocking spontaneous inhibitory events results in multiplicative downscaling of excitatory synaptic strength, dependent on transcription and BDNF signaling. These findings highlight the role of spontaneous inhibitory neurotransmission in setting excitatory synaptic strength at rest.
