Glucose transporters (GLUTs): Underreported yet crucial molecules in unraveling testicular toxicity

From oxidative imbalance to compromised standard sperm parameters: Toxicological aspect of phthalate esters on spermatozoa

Summary: The exponential rise in global male infertility and subfertility-related issues due to phthalate esters usage raises severe concern. This review focuses on the spermiotoxic effects of phthalate esters, discussing their impact on in vivo and in vitro models as well as surveyed human populations. The imbalance in pro-oxidant:anti-oxidant ratio caused by phthalates and subsequent loss of sperm structural and functional integrity are also critically discussed. Future research directions to address the knowledge gaps in phthalate-mediated male reproductive toxicity are provided.

Enhanced autophagy reversed aflatoxin B1-induced decrease in lactate secretion of dairy goat Sertoli cells

Summary: This study found that aflatoxins can disrupt the glutamine-lactate metabolism that supports the development of germ cells in dairy goat. It inhibits lactate secretion. Meanwhile, activated autophagy partially reverses the decrease in lactate secretion induced by aflatoxins by promoting glucose utilization and increasing the expression of proteins related to lactate secretion.

It shows that peroxisomal beta-oxidation plays a significant role in liver cholesterol biosynthesis by generating free acetate as a precursor. It suggests that induction of peroxisomal beta-oxidation serves as a mechanism for upregulation in cholesterol biosynthesis and contributes to diabetes-induced hypercholesterolemia.

Asprosin promotes steroidogenesis and spermatogenesis with improved glucose metabolism in adult mice testis

Summary: The study reveals that Asprosin plays a regulatory role in mouse testicular functions. It increases glucose and lactate levels, promotes cell proliferation and survival, and reduces germ cell apoptosis, resulting in increased sperm counts. Asprosin also facilitates steroidogenesis and spermatogenesis by promoting glucose uptake and lactate synthesis. The significant correlation between Asprosin-induced increased insulin receptor expression and increased testosterone, glucose, and lactate levels suggests its role in increased survival and proliferation but decrease in germ cell apoptosis. This study highlights the importance of Asprosin in regulating adult mouse testicular functions.

Inhibition of glucose transport synergizes with chemical or genetic disruption of mitochondrial metabolism and suppresses TCA cycle-deficient tumors

Summary: In this study, researchers developed a series of potent, orally bioavailable inhibitors of class I glucose transporters (GLUTs). The representative compound KL-11743 specifically blocks glucose metabolism, leading to an increase in oxidative phosphorylation in the mitochondria. Disrupting mitochondrial metabolism in combination with KL-11743 causes synthetic lethality, and patient-derived xenograft models demonstrate the vulnerability of succinate dehydrogenase A (SDHA)-deficient cancers to GLUT inhibitors.
