Design, synthesis, and molecular modeling of heterodimer and inhibitors of α-amylase as hypoglycemic agents

L-Arginine inhibits the activity of α-amylase: Rapid kinetics, interaction and functional implications

Summary: This study investigated the rapid unfolding kinetics and interaction mechanism of pancreas alpha-amylase (PPA) induced by L-arginine. The interaction between L-arginine and PPA resulted in a decrease in beta-sheet and an increase in random coil, leading to a decrease in enzyme activity. However, the unfolding of PPA could be compensated and the secondary structure change could be partially recovered by a macromolecular crowded medium. The results provide new insights into the interaction of L-arginine with digestive enzymes and the unfolding mechanism of enzymes.

Summary: This study investigated the protective effect of citric acid as a common food preservative against the formation of advanced glycation end-products (AGEs) and its interaction with α-amylase enzyme. The results showed that citric acid inhibited α-amylase activity and suppressed AGEs formation. Citric acid interacted with α-amylase and induced conformational changes. These findings are important for the management of type 2 diabetes complications.

Summary: The synthesized 3,3-di(indolyl)indolin-2-ones exhibited higher α-glucosidase inhibitory activities and lower α-amylase inhibitory activities. Compound 1i showed the most favorable inhibitory activity. Docking studies further supported the observed inhibitory activities. These results suggest that 3,3-di(indolyl)indolin-2-ones may serve as suitable α-glucosidase inhibitors, and the lower α-amylase activity could reduce side effects.
