Genetic ablation of diabetes-associated gene Ccdc92 reduces obesity and insulin resistance in mice

Lu Ren et al.

Summary: Multiple genome-wide association studies (GWAS) have identified specific genetic variants in the CCDC92 locus associated with obesity and type 2 diabetes in humans. This study demonstrates that knockout of the CCDC92 gene in mice reduces obesity and improves insulin sensitivity, protects against adipose tissue dysfunction, and attenuates hepatic steatosis. Furthermore, CCDC92 deletion inhibits inflammation and suppresses the NLRP3 inflammasome in white adipose tissue, suggesting CCDC92 as a potential target for treating obesity and insulin resistance.

The G protein-coupled oestrogen receptor GPER in health and disease: an update

Eric R. Prossnitz et al.

Summary: Oestrogens and their receptors have a significant impact on physiology and diseases, including the protection against various diseases in women. GPER, as a 7-transmembrane G protein-coupled receptor, plays a role in rapid signaling and transcriptional regulation. The research on GPER is important for understanding physiology, diseases, and potential therapeutic targets and prognostic indicators.

Myeloid-derived growth factor deficiency exacerbates mitotic catastrophe of podocytes in glomerular disease

Ping Zhan et al.

Summary: MYDGF plays an important role in podocytes by protecting against mitotic catastrophe and regulating the expression of the transcription factor RUNX2. Its reduction in glomeruli from patients with glomerular disease is correlated with glomerular filtration rate, serum creatinine, and podocyte loss.

Loss of CLDN5 in podocytes deregulates WIF1 to activate WNT signaling and contributes to kidney disease

Hui Sun et al.

Summary: Although mature podocytes lack tight junctions, the tight junction integral membrane protein claudin-5 (CLDN5) is predominantly expressed on the plasma membranes of podocytes under normal conditions. The deletion of CLDN5 exacerbates podocyte injury and proteinuria in a diabetic nephropathy mouse model by reducing ZO1 expression, inducing nuclear translocation of ZONAB, and downregulating the expression of WNT inhibitory factor-1 (WIF1).

The Updates of Podocyte Lipid Metabolism in Proteinuric Kidney Disease

Yu Sun et al.

Summary: Disorder of podocyte lipid metabolism is closely associated with various proteinuric kidney diseases, excessive lipid accumulation can lead to cellular dysfunction, ultimately resulting in cell hypertrophy, detachment, and death. Targeting podocyte lipid metabolism may represent a novel therapeutic strategy for patients with proteinuric kidney disease.

The ABCs of Sterol Transport

Ashlee M. Plummer et al.

Summary: Cholesterol homeostasis and trafficking are crucial for maintaining the asymmetric plasma membrane of eukaryotic cells, and disruptions in cholesterol trafficking can lead to various human diseases. ABC transporters, such as ABCA1 and ABCG5/G8, play key roles in this process, with mutations in these proteins linked to disorders like Tangier disease and sitosterolemia. These two proteins function in the reverse cholesterol transport pathway, mediating the efflux of cholesterol and xenosterols to high-density lipoprotein and bile salt micelles, highlighting the importance of understanding sterol transport driven by ABC transporters.
