Evaluation of chromogenic substrates for horseradish peroxidase on paper-based microfluidic devices

Summary: Microfluidic paper-based analytical devices (microPADs) are a promising platform for point-of-care assays. This study evaluated and compared 33 chromogenic substrate solutions for horseradish peroxidase (HRP) in terms of their analytical performance. The findings of this study will guide the selection and formulation of substrate solutions for future paper-based assays involving HRP.

Single-atom Rh nanozyme: An efficient catalyst for highly sensitive colorimetric detection of acetylcholinesterase activity and adrenaline

Summary: In this study, a single-atom nanozyme-Rh-N/C with tetra-enzyme mimic activities was constructed. The Rh-N/C showed stable and reusable laccase and oxidase mimic activities, even after 2 months of storage and 10 times of reuse with minimal loss of activity (maintained above 96.0%). Two biosensors were designed based on the versatile enzymatic activities of Rh-N/C, including a colorimetric biosensor for acetylcholinesterase activity determination and a sensitive Rh-N/C-based biosensor for adrenaline detection. This work expands the application of single-atom nanozyme and explores their potential in biosensors.

A 3D microfluidic paper-based analytical device with smartphone-assisted colorimetric detection for iodine speciation in seaweed samples

Summary: This study presents the development of a 3D origami microfluidic paper-based analytical device (3D mu PAD) for the determination of iodide and iodate in edible seaweeds by colorimetric detection using a smartphone. The addition of a methacrylate holder improved the interlayer contact in 3D mu PADs, resulting in enhanced sensitivity and precision, as well as real-time monitoring. The assay relies on the formation of a blue triiodide-starch complex at the detection areas of the 3D mu PAD, providing an affordable, fast, and environmentally friendly method for simultaneous determination of inorganic iodine species. Under optimal conditions, the method achieved low limits of detection and quantification for iodide and iodate.

Discovery and biosynthesis of karnamicins as angiotensin converting enzyme inhibitors

Summary: This study characterizes six karnamicins with fully substituted hydroxypyridine and thiazole moieties from the rare actinobacterium Lechevalieria rhizosphaerae NEAU-A2. Enzyme colorimetric cellulose membrane bioactivity strips based on acetylcholinesterase immobilization for inhibitors preliminary screening

Summary: To screen active pharmaceutical ingredients used as acetylcholinesterase inhibitors (AChEIs) for Alzheimer's disease treatment, a colorimetric cellulose membrane bioactivity strip (CBS) was developed. The strip enabled rapid and simple screening of AChEIs based on the colorimetric method. Inhibitors decreased the enzyme activity of AChE, resulting in decreased color intensity. The CBS exhibited fast chromogenic time, excellent specificity, and storage stability, making it a promising tool for preliminary screening of AChEIs and inspiring the development of other cellulose membrane-based biosensors.

Structure-Activity Relationships Reveal Beneficial Selectivity Profiles of Inhibitors Targeting Acetylcholinesterase of Disease-Transmitting Mosquitoes

Summary: Insecticide resistance poses a threat to the prevention of infectious diseases by controlling disease-transmitting mosquitoes. New insecticidal compounds with minimal adverse effects on humans and the environment are urgently required. In this study, we investigate noncovalent inhibitors of acetylcholinesterase (AChE), a well-established target for insecticides, and find that certain compounds based on a 4-thiazolidinone scaffold effectively inhibit AChE from Anopheles gambiae and Aedes aegypti mosquitoes. The selectivity of these compounds depends on the substitution pattern of the phenyl ring.
