Characterising heart rhythm abnormalities associated with Xp22.31 deletion

Georgina Wren et al.

Summary: This study found that genetic deletions at Xp22.31 are associated with an increased risk of abnormal heart rhythms (AHRs) and identified risk factors and comorbidities associated with AHRs. The study also found that deletion carriers overwhelmingly support the implementation of cardiac screening.

Naringenin in Si-Ni-San formula inhibits chronic psychological stress-induced breast cancer growth and metastasis by modulating estrogen metabolism through FXR/EST pathway

Juping Zhang et al.

Summary: This study aimed to explore the modulatory mechanism and bioactive compound of Si-Ni-San (SNS) in regulating estrogen metabolism during breast cancer development induced by chronic psychological stress. The results showed that SNS can inhibit breast cancer growth and metastasis by regulating the FXR/EST signaling pathway and promoting estradiol metabolism.

Hepatocyte estrogen sulfotransferase inhibition protects female mice from concanavalin A-induced T cell-mediated hepatitis independent of estrogens

Jingyuan Wang et al.

Summary: Autoimmune hepatitis (AIH) is a chronic liver disease with a higher incidence in females, and the molecular mechanism for female predisposition is poorly understood. Estrogen sulfotransferase (Est) was found to be highly induced in the liver of AIH mice, and inhibiting Est protected female mice from AIH regardless of estrogen levels. Mechanistically, Est ablation upregulated lipocalin 2 (Lcn2), and Lcn2 was found to be necessary for the protective effect of Est ablation.

Steroid Sulfation in Neurodegenerative Diseases

Jana Vitku et al.

Summary: This article focuses on the balance of steroid sulfation and desulfation in Alzheimer's disease, Parkinson's disease, and multiple sclerosis. Steroids in the brain and periphery have mutual influence, and play various roles in the nervous system, including effects on neurons and anti-inflammatory actions. The processes of sulfation and desulfation are crucial for the water solubility and transport of steroids.

Estrogen as a key regulator of energy homeostasis and metabolic health

Fatemeh Mahboobifard et al.

Summary: This article reviews the current understanding of estrogen's role in regulating energy balance at central and peripheral levels. It discusses the importance of estrogen signaling through nuclear and extranuclear pathways, the influence of sex differences on metabolic disorders, and the clinical significance of estrogen signaling in abnormal metabolic conditions such as obesity or high-fat diets. The review highlights research gaps in our understanding of metabolic diseases and their potential impact on developing more effective treatment options.

Steroid hormones sulfatase inactivation extends lifespan and ameliorates age-related diseases

Mercedes M. Perez-Jimenez et al.

Summary: The study demonstrates that loss of function of the steroid sulfatase sul-2 in Caenorhabditis elegans increases lifespan and ameliorates protein aggregation diseases, through mechanisms involving factors related to germline-mediated longevity. Sul-2 is predominantly expressed in sensory neurons, suggesting regulation by environmental cues.

Estrogen Sulfotransferase (SULT1E1): Its Molecular Regulation, Polymorphisms, and Clinical Perspectives

MyeongJin Yi et al.

Summary: Estrogen sulfotransferase (SULT1E1) is an important enzyme involved in sulfating estrogens, thyroid hormones, and certain medications. Genetic variants of SULT1E1 have been linked to diseases, especially sex-hormone-related cancers. Understanding the role and polymorphisms of SULT1E1 is crucial for its clinical relevance.

Summary: The STS enzyme plays a crucial role in the metabolism of steroid sex hormones and is associated with the pathophysiology of certain diseases. Successful STS inhibitor candidates like Irosustat have shown promise in treating specific cancers and neurodegenerative disorders, with reduced estrogenic side effects. Dual-acting molecules that inhibit STS and another synergistic mechanism may offer therapeutic efficiency.

Sex-specific expression mechanism of hepatic estrogen inactivating enzyme and transporters in diabetic women

Muluneh Fashe et al.

Summary: In menopause, circulating estrogen levels decrease significantly and the liver represses the capability of inactivating and excreting estrogens. Women are more prone to developing type 2 diabetes and liver diseases during this period. Phosphorylated nuclear receptors play a role in increasing the expression of genes involved in estrogen inactivation and excretion, possibly serving as a defense mechanism against estrogen toxicity in the livers of women.

The molecular basis of OH-PCB estrogen receptor activation

Summary: The molecular basis of OH-PCB inhibition of SULT1E1 is revealed for the first time, providing a novel template for designing high-affinity inhibitors. Mutants resistant to OH-PCBs weaken its affinity without affecting the enzyme's catalytic parameters, demonstrating that OH-PCBs regulate estrogen receptors by binding the OH-PCB binding pocket of SULT1E1.
