A Multiple Proton Transfer Mechanism for the Charging Step of the Aminoacylation Reaction at the Active Site of Aspartyl tRNA Synthetase

Saheb Dutta et al.

Summary: This study investigates the multiple proton transfer mechanism in the charging step of Aspartyl-tRNA synthetase catalysis through three QM/MM simulations. The most feasible pathway at the enzyme's active site is identified. The results provide important insights into the recognition and mechanism of the charging step.

Molecular Insights on Solvent Effects in Organic Reactions as Obtained through Computational Chemistry Tools

Manajit Das et al.

Summary: In this paper, the molecular understanding of the role of protic solvents in organic transformations is explored using density functional and ab initio computational studies. The inclusion of explicit solvent molecules in crucial transition states has been found to improve the energetic estimates of organocatalytic and transition-metal-catalyzed reactions. An overview of the importance of an explicit-implicit solvation model is provided with the help of interesting examples.

Beyond Continuum Solvent Models in Computational Homogeneous Catalysis

Gantulga Norjmaa et al.

Summary: The use of solvent effects in computational modeling of homogeneous catalysis is essential for capturing accurate mechanistic details. While continuum solvent models are commonly used, explicit solvent models are often necessary to provide a more detailed and accurate representation, leading to improved mechanistic understanding of reactions. Through selected examples, this article highlights the limitations of continuum solvent models and the benefits of incorporating explicit solvent molecules in quantum mechanical descriptions.

Solvent-Assisted Ketone Reduction by a Homogeneous Mn Catalyst

Annika M. Krieger et al.

Summary: This study investigates the impact of solvation on the mechanism of ketone reduction and uncovers a new possible mechanism. This mechanism does not require the participation of N-H moieties in manganese catalysts and is strongly influenced by the presence of solvent molecules. The results also demonstrate that the enantioselectivity induced by chiral ligands is maintained in this mechanism.

Reactivity and Enantioselectivity in NHC Organocatalysis Provide Evidence for the Complex Role of Modifications at the Secondary Sphere

Zayed Alassad et al.

Summary: Secondary-sphere interactions have been used to control reactivity and selectivity in organometallic and enzymatic catalysis. This study investigates the mechanistic pathway promoted by secondary-sphere modification in organocatalysis using boronic acids as modifiers. The results indicate stable off-cycle catalyst aggregation and BA-base adducts, and a rate-limiting, water-assisted hydrogen atom transfer mechanism. The presence of secondary-sphere modifiers also leads to an uncommon Curtin-Hammett-controlled enantioselectivity.

Free Energy Landscape of the Adenylation Reaction of the Aminoacylation Process at the Active Site of Aspartyl tRNA Synthetase

Saheb Dutta et al.

Summary: This study investigates the adenylation reaction between aspartic acid and ATP at the active site of AspRS in Escherichia coli through QM/MM simulations. The results show that the mutation of a conserved residue at the active site increases the free energy barrier of the reaction. Additionally, the study provides molecular details of structural changes during the reaction.

Phosphine-Free Manganese Catalyst Enables Selective Transfer Hydrogenation of Nitriles to Primary and Secondary Amines Using Ammonia-Borane

Koushik Sarkar et al.

Summary: Primary and secondary amines were synthesized via nitrile hydrogenation using a borrowing hydrogenation strategy catalyzed by phosphine-free manganese complexes. The method is chemodivergent, allowing control over the final product by adjusting catalyst structure and reaction polarity. The protocol operates without the need for externally added base or high-pressure dihydrogen gas, providing a practical and efficient route to a variety of amines.
