Genetic diversity and detection of atypical porcine pestivirus infections

Summary: This study revealed that the 5' untranslated region (5' UTR) of atypical porcine pestivirus (APPV) has a higher degree of sequence conservation, and some highly conserved segments of the 5' UTR overlap with important functional regions in viral protein translation. A newly designed qPCR assay targeting the conserved 5' UTR regions was able to detect APPV in well-documented cases of congenital tremor from multiple farm sites in the U.S. Midwest.

Genetic characterization of atypical porcine pestivirus from neonatal piglets with congenital tremor in Hubei province, China

Summary: This study determined the genetic characteristics of APPV in Hubei province, China. The results showed that APPV mainly caused histological lesions in the central nervous system and immune tissues of CT-affected piglets. Furthermore, phylogenetic analysis revealed the close relationship between the CH-HB2020 and CH-HB2021 strains and APPV_CH-GX2016. These findings enhance our understanding of APPV pathogenesis and provide potential molecular evidence for its prevalence and transmission.

The Diversity and Spatiotemporally Evolutionary Dynamic of Atypical Porcine Pestivirus in China

Summary: This study investigates the source, evolutionary history, and transmission pattern of atypical porcine pestivirus (APPV) in China. Phylogenetic analyses reveal four distinct lineages of APPV in China, with the greatest diversity observed in this country. Bayesian analyses suggest that the E2 gene of APPV evolves at a mean rate of 1.22 x 10(-3) substitutions/site/year. The most recent common ancestor of APPV is estimated to have existed in 1886, earlier than the emergence of congenital tremor (CT) in 1922. The study also finds that APPV possibly originated in the Netherlands and was introduced into China between 1837 and 2010. Guangdong, together with central and southwest China, played a significant role in the dispersal of APPV within China. The transmission pattern of China lineages shows a south to north movement tendency, possibly influenced by strict environmental policies implemented in China since 2000. The findings also suggest that the diversification of APPV lineages is driven by positively selected pressure and geographic adaptations.

Evolution and genetic diversity of atypical porcine pestivirus (APPV) from piglets with congenital tremor in Guangxi Province, Southern China
