Summary: The study characterized the damage caused by sodium iodate (NaIO3) in mouse retinal pigment epithelium (RPE), which was divided into three regions: periphery with normal-shaped RPE, transitional zone with elongated cells, and center with severely damaged or lost RPE. Elongated cells in the transitional zone displayed molecular characteristics of epithelial-mesenchymal transition. Central RPE was more susceptible to stresses than peripheral RPE. Additionally, SIRT6, a protein deacylase, depletion was observed in the nucleus under stress, which could be overcome by SIRT6 overexpression in transgenic mice, protecting RPE from NaIO3-induced damage.

NaIO3 administration results in the disappearance of RPE marker OTX2 from the central RPE, the inability to identify choroid capillaries adjacent to RPE, and the destruction of choroid capillaries. Additionally, the expression level of Vegfa in RPE decreases rapidly after NaIO3 administration.

Summary: The dry form of age-related macular degeneration (AMD) is a major cause of vision loss. A study found that STING and cGAS expression is increased in the macular retinas of dry AMD patients. The activation of cGAS-STING was also observed in oxidative stress-induced mouse retina degeneration, and treatment with the BRD4 inhibitor JQ1 reduced this activation and alleviated inflammation and photoreceptor degeneration.

Summary: The lack of suitable animal models for (dry) age-related macular degeneration (AMD) has hindered therapeutic research into the disease. In this study, pigmented rats and mice were systematically injected with various doses of sodium iodate (SI) to evaluate their suitability as models for studying experimental therapies for retinal degenerative diseases. The results showed dose-dependent structural and functional pathological effects on the retinal pigment epithelium (RPE) and retina, with a dose of 30 mg/kg appearing to be suitable for future studies.
