Non-invasive treatment of ischemia/reperfusion injury: Effective transmission of therapeutic near-infrared light into the human brain through soft skin-conforming silicone waveguides

Paul T. Morse et al.

Summary: Noninvasive delivery of near-infrared light (IRL) to human tissues has shown promising results for the treatment of acute and chronic diseases. In this study, we developed IRL delivery waveguides (IDWs) that allow efficient and safe delivery of IRL to the brain. The IDWs have unique design features to ensure uniform IRL delivery and prevent skin burns. Through testing on human cadavers and isolated tissues, we found that IDWs outperformed fiberoptic cables in IRL transmission. This study discusses the potential of IDWs for future clinical use and suggests further improvements.

Photobiomodulation of Cytochrome c Oxidase by Chronic Transcranial Laser in Young and Aged Brains

Fabrizio dos Santos Cardoso et al.

Summary: This study provides the first detailed brain mapping of the effects of chronic photobiomodulation (PBM) on cellular cytochrome c oxidase (CCO) activity, as well as the long-term effects of PBM on young and aged brains. The findings suggest that aging decreases regional brain CCO activity and functional connectivity, while chronic laser stimulation reverses these age-related effects.

Sometimes less is more: inhibitory infrared light during early reperfusion calms hyperactive mitochondria and suppresses reperfusion injury

Paul T. Morse et al.

Summary: Ischemic stroke is a global disease that affects over 77 million people annually. Mitochondria play a crucial role in early ischemia-reperfusion injury, contributing to the generation of reactive oxygen species and damage to neuronal tissue. This review explores the relationship between mitochondrial membrane potential and ROS production, as well as the impact of serine 47 dephosphorylation of cytochrome c during ischemia. The use of near infrared light to treat stroke is also discussed, highlighting the importance of selecting the correct wavelength for optimal outcomes.

Mitochondrial fission and mitophagy are independent mechanisms regulating ischemia/reperfusion injury in primary neurons

Anthony R. Anzell et al.

Summary: Mitochondrial dynamics and mitophagy are essential for maintaining a healthy mitochondrial network, and their disruption can lead to mitochondrial dysfunction. Research suggests that these systems are interrelated, but their specific roles in ischemia/reperfusion injury are still unclear. Using a mouse model and evaluating mitochondrial morphology and mitophagic flux, it was found that Drp1 plays a key role in the progression of I/R injury, while mitophagy can occur independently of Drp1-mediated mitochondrial fission.

Hypothermia versus Normothermia after Out-of-Hospital Cardiac Arrest

Josef Dankiewicz et al.

Summary: A randomized trial comparing hypothermia and normothermia treatment after out-of-hospital cardiac arrest showed no significant differences in death or functional outcomes at 6 months. The hypothermia group had a higher incidence of arrhythmia resulting in hemodynamic compromise compared to the normothermia group.

Transcranial Near Infrared Light Stimulations Improve Cognition in Patients with Dementia

Damir Nizamutdinov et al.

Summary: The study examined the safety and therapeutic benefits of transcranial near-infrared (tNIR) light stimulations in treating dementia. The active treatment group showed improvements in cognitive functions, sleep quality, and mood, suggesting potential benefits of tNIR light therapy for dementia patients. This study supports the need for further trials focusing on alleviating caregivers' burden in dementia treatment.

Cytochrome c oxidase-modulatory near-infrared light penetration into the human brain: Implications for the noninvasive treatment of ischemia/reperfusion injury

Paul T. Morse et al.

Summary: Research has shown that specific wavelengths of near-infrared light can protect against brain ischemia/reperfusion injury in rodents by inhibiting mitochondrial enzymes, with the light also penetrating through cadaver heads to suggest potential therapeutic application for human brain injury, regardless of skin pigmentation.
