Therefore, the development of ONOO--activated near-infrared (NIR) fluorescent probes with high sensitivity and selectivity is crucial for early diagnosis of DILI.

Phenotyping of Methicillin-Resistant Staphylococcus aureus Using a Ratiometric Sensor Array

Summary: Chemical tools that can classify multidrug-resistant bacteria (superbugs) are helpful for early disease diagnosis and precise therapy. We introduce a sensor array that can easily phenotype methicillin-resistant Staphylococcus aureus (MRSA), a common superbug in clinical practice. The array consists of eight ratiometric fluorescent probes with characteristic vibration-induced emission (VIE) profiles, which interact with the cell walls of bacteria and produce varying fluorescence intensity ratios. The differences in these ratios result in fingerprints for different genotypes of MRSA, allowing their identification using principal component analysis (PCA) without the need for cell lysis and nucleic acid isolation. The results obtained with this sensor array are consistent with polymerase chain reaction (PCR) analysis.

The design of small-molecule prodrugs and activatable phototherapeutics for cancer therapy

Orthogonally Engineered Albumin with Attenuated Macrophage Phagocytosis for the Targeted Visualization and Phototherapy of Liver Cancer

Summary: We report a strategy based on engineered human serum albumin (HSA) that allows the targeted delivery of diagnostic and therapeutic agents for hepatocellular carcinoma (HCC). This approach involves covalent cysteine conjugation and host-guest chemistry to functionalize HSA with two peptides, one targeting an HCC-specific biomarker and the other reducing macrophage phagocytosis. The engineered HSA effectively delivered fluorescent and phototherapeutic agents to HCC, enabling target-specific visualization and photodynamic ablation in vivo.

Selective detection of peroxynitrite using an isatin receptor and a naphthalimide fluorophore

Summary: Peroxynitrite is a reactive oxygen and nitrogen species that plays a role in various biological reactions, making it crucial to detect and track it in biological systems. In this study, a new turn-on probe encapsulated in PEG DSPE-PEG/HN-I was used to rapidly fluorescently detect ONOO-. The encapsulation of HN-I using DSPE-PEG2000 optimized the sensing abilities of the naphthalimide probe and prevented ACQ. The use of DSPE-PEG/HN-I to detect changes in exogenous ONOO- levels in HepG2 cells and endogenous ONOO- induced by LPS in RAW 267.4 cells was demonstrated.

Dual-Channel Fluorescent Probe for the Simultaneous Monitoring of Peroxynitrite and Adenosine-5′-triphosphate in Cellular Applications

Summary: The fluorescent probe ATP-LW was developed for simultaneous detection of ONOO- and ATP levels. By utilizing different emission properties of the products, ONOO- levels can be monitored in the green channel and ATP concentrations in the red channel. The probe was successfully used in hepatocytes to demonstrate changes in signal intensity in response to ATP synthase inhibition and exogenous ONOO- exposure.
