In-Cell Chemical Crosslinking Identifies Hotspots for SQSTM-1/p62-IKBα Interaction That Underscore a Critical Role of p62 in Limiting NF-KB Activation Through IKBα Stabilization

Summary: We have found that the protein scaffold p62/SQSTM1 is closely associated with IκBα in liver cells, leading to the stabilization and nuclear entry of IκBα. This association is necessary for the termination of NF-κB activation cycle. Our study defines the structural hotspots of p62-IκBα association and reveals the influence of p62 on IκBα-protein interactions. We speculate that p62 may be involved in the piggy-back nuclear transport of IκBα following its NF-κB-elicited transcriptional activation.

Summary: This study analyzed the influence of the RelA TAD on the NFKB-DNA interactions, and found that the presence of the TAD increased the stoichiometry of NFKB-DNA complexes and enhanced the binding affinity of p50/RelA for DNA sequences. However, it also led to an increase in the affinity for nonspecific DNA sequences and a decrease in specificity for KB DNA sequences.

Single-molecule conformational dynamics of a transcription factor reveals a continuum of binding modes controlling association and dissociation

Summary: The binding and release of transcription factor NF-kappa B from DNA are kinetically controlled by structural dynamics, with a continuum of conformations interconverting between free and DNA-bound states. The inhibitor protein I kappa B alpha accelerates dissociation and induces a locked conformation where NF-kappa B cannot bind DNA. This study suggests a novel mechanism for controlling the association and dissociation of transcription factors through a continuum of binding modes.
