A novel theoretical strategy for predicting dissolution kinetics and mechanisms of pharmaceuticals in complex biorelevant media

The effect of buffer species on biorelevant dissolution and precipitation assays-Comparison of phosphate and bicarbonate buffer

Summary: Biorelevant solubility and dissolution testing is an important tool in pharmaceutical development. However, the use of biorelevant media does not always match the solubility data observed in human intestinal fluids. This study found that the medium primarily affects dissolution kinetics, and the unique buffering properties of bicarbonate buffer play a special role in simulating conditions in human intestinal fluids. Further investigation is needed to understand the factors causing differences in solubility and dissolution behavior between phosphate- and bicarbonate-buffered biorelevant media.

The role of hydrogen-bond in solubilizing drugs by ionic liquids: A molecular dynamics and density functional theory study

Summary: This study systematically investigated the solvation mechanisms of aspirin and etomidate in four combinations of ionic liquids through molecular dynamics simulations and DFT calculations. The results showed that the solvation properties were closely related to the hydrogen-bond basicity of anions and the hydrogen-bond acidity of cations. The study also found differences in solvation mechanisms between aspirin and etomidate.

Theoretical modeling and prediction of biorelevant solubility of poorly soluble pharmaceuticals

Similarity of dissolution profiles from biorelevant media: Assessment of interday repeatability, interanalyst repeatability, and interlaboratory reproducibility using ibuprofen and ketoconazole tablets

Summary: This study assessed the repeatability and reproducibility of dissolution profiles from biorelevant media, showing favorable performance in interday repeatability, interanalyst repeatability, and interlaboratory reproducibility. Commercial media exhibited greater interlaboratory reproducibility than 'from scratch' media, and biorelevant media were able to detect differences when products were not similar. Overall, biorelevant media demonstrated favorable repeatability and reproducibility performance.

Influence of excipients on thermodynamic phase behavior of pharmaceutical/solvent systems: Molecular thermodynamic model prediction

Summary: In this study, the influence of polymeric excipients on the thermodynamic phase behavior of active pharmaceutical ingredients and its aqueous solution was systematically investigated. The results showed that all four excipients could effectively solubilize the API, providing theoretical guidance for the screening of pharmaceutical formulation excipients. The thermodynamic phase behavior of the API in the excipients/water ternary system was accurately predicted by the PC-SAFT model, demonstrating good agreement with experimental results.

Insights into influence mechanism of polymeric excipients on dissolution of drug formulations: A molecular interaction-based theoretical model analysis and prediction

Summary: This study utilized a combination of PC-SAFT thermodynamic model and chemical-potential-gradient model to analyze the dissolution mechanism of poorly water-soluble drugs, IND and NAP, from PVAc and PVPVA 64 formulations. The results demonstrated that this combined approach is efficient in explaining the drug dissolution mechanism from these polymeric formulations.
