Control of API release with matrix polymorphism in tristearin microspheres

Summary: Glycerides are commonly used as solid matrices in pharmaceutical intermediates and dosage forms. The release of drugs from these matrices is controlled by diffusion-based mechanisms, with chemical and crystal polymorph differences of the lipid matrix influencing the release rates. This study focuses on the impacts of tristearin polymorphism on drug release using model formulations containing crystalline caffeine. The results show that the release rates are influenced by the polymorphic states of tristearin and the conversion routes between them. The findings provide valuable insights for formulators in understanding the impacts of triglyceride polymorphism on drug release rates.

Phase Behavior of Drug-Lipid-Surfactant Ternary Systems toward Understanding the Annealing-Induced Change

The observed higher miscibility of Kolliphor P 407 with specific components was proposed as the trigger for physical separation during melt solidification, with phase separation underlying the formation of a stable beta-polymorphic form of Compritol upon annealing.

Solid lipid microparticles loaded with ascorbic acid: Release kinetic profile during thermal stability

Liquid Lipids Act as Polymorphic Modifiers of Tristearin-Based Formulations Produced by Melting Technologies

Summary: This study evaluated the impact of different oral-approved liquid lipids on the polymorphism, phase transitions, and structure of solid lipid-based formulations, and their influence on drug release. The addition of liquid lipids was found to promote the transition of tristearin from the alpha-form to the stable beta-form, affecting the crystal structure of tristearin-based formulations and influencing the release behavior of a model hydrophilic drug. Overall, the addition of liquid lipids can be considered an interesting approach to control triglyceride crystallization in the beta-form, enabling the production of stable lipid formulations.
