1,3-Diamine-Derived Catalysts: Design, Synthesis, and the Use in Enantioselective Mannich Reactions of Ketones

Summary: 1,3-Diamine-derived catalysts were synthesized and utilized in asymmetric Mannich reactions of ketones. The Mannich products with high enantioselectivities were obtained under mild conditions through catalysis by one of the 1,3-diamine derivatives in the presence of acids. Bond formation primarily occurred at the less-substituted α-position of the ketone carbonyl group. The primary and tertiary amines of the 1,3-diamine derivative cooperatively acted for the catalysis.

Control of Reactions of Pyruvates by Catalysts: Direct Enantioselective Mannich Reactions of Pyruvates Catalyzed by Amine-based Catalyst Systems

Summary: This study reports amine-based catalyst systems for the enantioselective Mannich reactions of pyruvates as nucleophiles. The reactions of pyruvates and cyclic sulfonylimines yielded the desired Mannich products, including those with tetrasubstituted carbon centers, in high yields and enantioselectivities. The choice of acid in the amine-based catalyst system played a key role in the formation of Mannich products with high enantioselectivities.

CuH-Catalyzed Enantioselective Reductive Coupling of 1,3-Dienes and Trifluoromethyl Ketoimines or ?-Iminoacetates

Summary: In this study, the asymmetric cross reductive coupling reaction of conjugated dienes and ketoimines using copper hydride as a catalyst was achieved for the first time. The reaction, catalyzed by chiral Ph-BPE ligand, provides a rapid and efficient access to homoallylic amines with two vicinal stereogenic centers.

Organocatalytic Enantioselective γ-Position-Selective Mannich Reactions of β-Ketocarbonyl Derivatives

Summary: Catalytic asymmetric Mannich reactions of beta-ketocarbonyl derivatives, such as beta-ketoesters and (2-oxopropyl)-phosphonate, have been reported. These reactions result in the formation of C-C bonds at the gamma-position of the beta-ketocarbonyl derivatives with high enantioselectivities. The reversible bond formation at the alpha-position of the beta-ketoester and the stable formation of the gamma-position-reacted product delta-amino beta-ketoester derivative are attributed to the dynamic kinetic process under catalytic conditions.

One-Pot Catalytic Synthesis of α-Tetrasubstituted Amino Acid Derivatives via In Situ Generation of N-Unsubstituted Ketimines
