Oxygen availability influences the incidence of testicular teratoma in Dnd1Ter/+ mice

Ximena M. M. Bustamante-Marin et al.

Summary: Testicular teratomas and teratocarcinomas are frequently found unilaterally in the left testis, and a point mutation in the Dnd1 gene can lead to an increased incidence of bilateral tumors in mice. Exposure to acute low oxygen conditions during fetal development increases the occurrence of bilateral teratoma in male gonads. This is associated with the maintenance of pluripotency genes, elevated Nodal signaling pathway activity, and suppressed germ cell mitotic arrest.

Belzutifan: a novel therapy for von Hippel-Lindau disease

Jingcheng Zhou et al.

Summary: A recent clinical trial has shown promising efficacy and safety data for belzutifan in patients with von Hippel-Lindau (VHL) disease-associated renal cell carcinoma. Based on these results, belzutifan has become the first therapeutic agent approved for the systemic treatment of cancer associated with VHL disease.

A novel high throughput screen to identify candidate molecular networks that regulate spermatogenic stem cell functions(dagger)

Tessa Lord et al.

Summary: This study developed a high-throughput method to rapidly predict alterations in functional capacity of spermatogonia. By conducting a large-scale RNAi screen of transcription factors, a repository of new information was generated, from which candidate molecular networks regulating regenerative functions were predicted through pathway analysis. The SRCAP-CREBBP/EP300 complex was identified as a mediator of differential levels of histone acetylation between stem cell and progenitor spermatogonia, influencing the expression of key self-renewal genes.

Hypobaric hypoxia exposure alters transcriptome in mouse testis and impairs spermatogenesis in offspring

Shuang Li et al.

Summary: Hypoxia exposure negatively affects spermatogenesis, reducing the number of undifferentiated spermatogonia and increasing the percentage of abnormal sperm. Additionally, hypoxia alters testicular gene expression and leads to long-lasting damage to spermatogenesis.

Distinctive molecular features of regenerative stem cells in the damaged male germline

Hue M. La et al.

Summary: This study uncovers the roles of growth factor signalling and mTORC1 in the regeneration of spermatogonial stem cells (SSCs). Single-cell analysis reveals the unique molecular features of regenerative SSCs and the changes in the composition of undifferentiated spermatogonia during germline recovery. The data suggest that transient mTORC1 activation is critical for the regenerative response, while sustained mTORC1 signalling is detrimental for SSC maintenance. Inhibition of growth factor signalling disrupts the regenerative state and limits germline recovery.

An interplay of NOX1-derived ROS and oxygen determines the spermatogonial stem cell self-renewal efficiency under hypoxia

Hiroko Morimoto et al.

Summary: The origin of ROS and oxygen tension play a critical role in driving spermatogonial stem cell (SSC) self-renewal. Under hypoxia, NOX1-derived ROS significantly decrease, impacting the proliferation of SSCs.

Acrylamide modulates the mouse epididymal proteome to drive alterations in the sperm small non-coding RNA profile and dysregulate embryo development

Natalie A. Trigg et al.

Summary: Exposure to environmental stressors can impact the sperm sncRNA profile and have significant post-fertilization consequences. Research shows that sncRNAs responsive to the reproductive toxicant acrylamide accumulate differently in sperm as they pass through the caput segment of the epididymis, affecting gene expression and early embryonic development after fertilization.

Gross and microanatomy of the male reproductive duct system of the saltwater crocodile Crocodylus porosus

Brett Nixon et al.

Summary: This study investigated the morphology and histology of the male reproductive system of crocodiles, focusing on the role of the excurrent duct system in sperm production and modification. The findings suggest that the ducts are involved in seminal fluid production and/or modification, contributing to the nourishment, protection, and storage of crocodile sperm. The structural characteristics of the crocodile male excurrent duct system share closer similarities with birds than with other groups within the Reptilia class or Mammalia, reflecting their common Archosaurs ancestry.
