The transcriptomic response to cannabidiol of Treponema denticola, a phytocannabinoid-resistant periodontal pathogen

Localization and pathogenic role of the cysteine protease dentipain in Treponema denticola

Summary: The Msp protein complex and the serine protease dentilisin are the best-characterized virulence factors in Treponema denticola, the major etiological agent of chronic periodontitis. In addition, the cysteine protease dentipain also contributes to pathogenicity, but its secretion, processing, cellular localization, and role in T. denticola virulence are not fully understood. This study found that dentipain is located in the outer sheath derived from T. denticola dentilisin- and Msp-deficient mutants, suggesting that dentilisin and Msp are involved in the secretion and maturation of dentipain. Inactivation of the dentipain gene slowed the growth of T. denticola, and the mutant strain showed altered gene expression, increased hydrophobicity, reduced autoaggregation activity, and decreased adhesion to IgG.

Cannabidiol attenuates periodontal inflammation through inhibiting TLR4/NF-κB pathway

Summary: This study aims to investigate the anti-inflammatory and osteoprotective effects of CBD in periodontitis and its molecular mechanisms. The results show that CBD significantly inhibits bone loss in experimental rat periodontitis models and reduces LPS-induced inflammation in human periodontal ligament cells (hPDLCs) by inhibiting TLR4/NF-kappa B pathway activation. Therefore, CBD can be considered as a therapeutic agent for periodontitis.

Measuring the diversity gap of cannabis clinical trial participants compared to people who report using cannabis

Summary: Little information is available regarding the demographics of cannabis users and how usage trends have changed over time. This makes it difficult to determine if participants in cannabis clinical trials represent the broader population of cannabis users. To address this knowledge gap, data from the National Survey on Drug Use and Health (NSDUH) was analyzed to examine cannabis usage among different population subgroups in the United States from 2002 to 2021. The results revealed significant increases in past-month cannabis usage among those aged 65 and older (2,066.1%) and 50-64-year-olds (472.4%). Furthermore, analysis of participant demographics in cannabis clinical trials showed an overrepresentation of white males in their 20s and 30s, indicating a structural discrimination that perpetuates social and health inequities.

Association of oral bacteria with oral hygiene habits and self-reported gingival bleeding

Summary: Research findings have shown that increasing self-reported gingival bleeding frequency is associated with increased gingival bacterial diversity. Frequent gingival bleeding is associated with increased abundance of certain bacteria (such as Porphyromonas endodontalis, Treponema denticola, and Fretibacterium spp.), but decreased abundance of bacteria within the gram-positive phyla Firmicutes and Actinobacteria. Additionally, using dental floss and mouthwash twice daily is associated with increased total abundance of bacteria in the Proteobacteria phylum, but decreased bacterial diversity.

Microbiome profiles of non-responding and responding paired periodontitis sites within the same participants following non-surgical treatment

Summary: This study aimed to compare the microbiome of sites that did not respond to non-surgical debridement treatment with those that responded well. The results showed no significant differences in clinical parameters and microbial communities between good responding and non-responding sites before and after treatment. In non-responding sites, the pathogenic bacterial community was not disrupted by the treatment.

Summary: Periodontal disease is primarily driven by dysbiosis in the oral microbiome, leading to the overrepresentation of species that induce pro-inflammatory responses in the periodontium. The oral spirochete Treponema denticola is consistently found at significantly elevated levels in periodontal lesions and is associated with the activation of tissue-destructive genes, mainly through the surface-expressed protease complex dentilisin, stimulating tissue-destructive cellular processes in a TLR2/MyD88/Sp1-dependent fashion. These findings provide new insights into the molecular mechanisms underlying periodontal disease progression and could potentially lead to the development of more effective therapeutic treatments.

Analyses of the Effects of Arginine, Nicotine, Serotype and Collagen-Binding Proteins on Biofilm Development by 33 Strains of Streptococcus mutans
