Validation of a pharmacological imaging challenge using 11C-buprenorphine and 18F-2-fluoro-2-deoxy-D-glucose positron emission tomography to study the effects of buprenorphine to the rat brain

Amelie Soyer et al.

Summary: The study used Buprenorphine and F-18-FDG PET imaging in isoflurane anesthetized rats to investigate the CNS effects of full receptor occupancy by this partial mu-OR agonist. The method was not found to be more sensitive in awake animals. This strategy may be useful for studying mu-OR desensitization associated with opioid tolerance in vivo.

Shared Mechanisms of GABAergic and Opioidergic Transmission Regulate Corticolimbic Reward Systems and Cognitive Aspects of Motivational Behaviors

Oveis Hosseinzadeh Sahafi et al.

Summary: The functional interplay between corticolimbic GABAergic and opioidergic systems is crucial for regulating reward and cognitive aspects of motivational behaviors, leading to the development of addictive behaviors and disorders. This review provides a comprehensive understanding of the shared mechanisms of GABAergic and opioidergic transmission, focusing on their modulation of dopaminergic activity in the ventral tegmental area (VTA), a central hub of the reward mechanisms.

Buprenorphine: a treatment and cause of opioid-induced respiratory depression

Albert Dahan et al.

Summary: Buprenorphine, a partial agonist at the mu opioid receptor, is considered relatively safe due to its low maximum effect on respiratory depression. However, when combined with sedatives like benzodiazepines, it can cause serious respiratory depression.

A local-neighborhood Lassen plot filter for creating occupancy and non-displaceable binding images

Bart de Laat et al.

Summary: This study introduces a new method, the Lassen plot filter, for estimating receptor occupancy of an exogenous drug in PET images, addressing the issue of spatial variation of ODrug. Simulated data and a human [C-11]flumazenil occupancy study demonstrated that the Lassen plot filter provides unbiased estimates in the presence of regional variation in VND or ODrug.

Complete inhibition of ABCB1 and ABCG2 at the blood-brain barrier by co-infusion of erlotinib and tariquidar to improve brain delivery of the model ABCB1/ABCG2 substrate [11C]erlotinib

Nicolas Tournier et al.

Summary: This study investigated the feasibility of simultaneous inhibition of ABCB1 and ABCG2 at the blood-brain barrier by co-infusion of erlotinib and tariquidar. The combination led to increased brain distribution of the model substrate in mice and macaques, potentially enhancing brain delivery of molecularly targeted anticancer drugs for the treatment of brain tumors. Additionally, tolerability and induction of apoptosis were assessed in human cerebral organoids, showing promising results for future applications.

Pharmacokinetic neuroimaging to study the dose-related brain kinetics and target engagement of buprenorphine in vivo

Sylvain Auvity et al.

Summary: A wide range of buprenorphine doses are used for either pain management or maintenance therapy in opioid addiction. The complex in vitro profile of buprenorphine, with affinity for mu-, delta-, and kappa-opioid receptors, makes it difficult to predict its dose-related neuropharmacology in vivo. PET imaging and kinetic modeling enabled description of the regional brain kinetics of C-11-buprenorphine with varying doses, revealing regional differences in buprenorphine-associated receptor occupancy. This strategy could be further combined with other techniques to investigate the neuropharmacokinetics and neuroreceptor correlate, particularly at mu-opioid receptors, of the acute effects of buprenorphine in humans.

Neuronal mechanisms underlying opioid-induced respiratory depression: our current understanding

Jan-Marino Ramirez et al.

Summary: Opioid-induced respiratory depression is the primary cause of death associated with therapeutic and recreational opioid use, with the death rate from opioid abuse growing disproportionally in the United States since the early 1990s. Despite uncovering many fundamental cellular and systems-level mechanisms related to opioid-related death, factors such as individual vulnerability, neuromodulatory compensation, and redundancy of opioid effects continue to hinder the formation of a concise, integrative view of OIRD. This review brings together multiple perspectives in the field of OIRD to create a comprehensive understanding of this important research topic and its future direction.

Positive allosteric modulation of the mu-opioid receptor produces analgesia with reduced side effects

Ram Kandasamy et al.

Summary: Positive allosteric modulators (PAMs) of the mu-opioid receptor (MOR) have been suggested as potentially safer analgesics compared to traditional opioid drugs, enhancing the activity of endogenous opioid peptides while reducing side effects. Experimental evidence shows that a specific mu-PAM, BMS-986122, can enhance the potency of endogenous opioid peptides and produce antinociceptive effects in mouse models of acute and inflammatory pain. This innovative approach to pain management may offer a safer alternative to conventional opioid medications.
