Soluble adenylyl cyclase contributes to imiquimod-mediated inflammation and is a potential therapeutic target in psoriasis

Jaewon You et al.

Summary: This study demonstrates the importance of soluble adenylate cyclase (sAC) in psoriasis pathogenesis and identifies it as a potential therapeutic target. The absence of Adcy10 gene significantly reduces the symptoms of psoriasiform dermatitis in mice and inhibits the differentiation and gene expression of Th17 cells. Moreover, topical application of sAC inhibitors shows promise in reducing psoriasiform dermatitis and IL17 gene expression in the skin, suggesting their potential as topical therapeutics for Th17-mediated skin diseases.

Cholangiocytes express an isoform of soluble adenylyl cyclase that is N-linked glycosylated and secreted in extracellular vesicles

Simei Go et al.

Summary: Soluble adenylyl cyclase (sAC)-derived cAMP is regulated by glucose levels, and N-linked glycosylated sAC isoforms are released into extracellular vesicles. Maturation and trafficking of sAC in cholangiocytes involves distinct sugar chains and vesicular machinery. The turnover of intracellular sAC(85) is rapid, with release of sAC(EV) detected within 3 hours.

Immunologic Responses and the Pathophysiology of Primary Biliary Cholangitis

Ruiling Chen et al.

Summary: Current understanding of the etiopathogenesis of PBC suggests that immune dysregulation plays a central role in the excessive immune responses against biliary epithelial cells. This is influenced by genetic background, environmental triggers, and epigenetic alterations. PBC is an autoimmune disease, and the contribution of the X chromosome in its pathogenesis remains unclear. Changes in the composition and function of the gut microbiome are closely related to PBC, but whether dysbiosis of the microbiome is a cause or a consequence of PBC pathogenesis is still ambiguous.

Primary biliary cholangitis: pathogenic mechanisms

Jesus Prieto et al.

Summary: PBC is characterized by autoimmune damage of intrahepatic bile ducts, with a higher prevalence in females. Recent findings suggest that PBC may be associated with dysregulation of mitophagy, AE2 deficiency, and altered immunoregulation.

Soluble adenylyl cyclase regulates the cytosolic NADH/NAD+ redox state and the bioenergetic switch between glycolysis and oxidative phosphorylation

Jung-Chin Chang et al.

Summary: Inhibiting sAC leads to increased extracellular lactate production and decreased pyruvate production, reflecting altered cytosolic NADH/NAD(+) ratio. sAC inhibition specifically suppresses complex I of the mitochondrial respiratory chain, while activating Epac1 of cAMP signaling increases cytosolic NADH/NAD(+) ratio, regulating cellular oxidative phosphorylation and glycolysis to maintain energy balance within the cell.

Cyclic AMP-binding protein Epac1 acts as a metabolic sensor to promote cardiomyocyte lipotoxicity

Marion Laudette et al.

Summary: The study reveals that dietary saturated fatty acid palmitate increases cAMP synthesis, disrupting lipid metabolism balance in cardiomyocytes and leading to mitochondrial dysfunction and ultimately cardiomyocyte death. Epac1 plays a crucial role in this process by modulating mitochondrial FA uptake and oxidation, with potential implications in cardiac pathophysiology.
