Glycan Node Analysis Detects Varying Glycosaminoglycan Levels in Melanoma-Derived Extracellular Vesicles

Summary: Extracellular vesicles (EVs) are important in mediating cell communication in (patho)physiological processes. However, the analysis of glycans and glycosaminoglycans (GAGs) in EVs has been overlooked due to technical challenges. In this study, a tangential flow filtration-based EV isolation method was coupled with glycan node analysis (GNA) to comprehensively analyze the glyco-polymer features of EVs. GNA identified EV-associated glyco-polymers that couldn't be detected with conventional mass spectrometry methods, providing valuable insights into the EV glycocode and its biological functions.

Vasculature organotropism in drug delivery

Summary: In recent decades, there has been a significant increase in the development of nanodelivery systems for preclinical and clinical applications, particularly in delivering RNA and protein-based therapeutics. The specific vasculature of organs provides a promising avenue for targeted delivery of nanoparticles and extracellular vesicles to interstitial cells. This article emphasizes the properties of organ-specific vasculature, the strategies for nanodelivery that exploit the organotropism of vasculature, and the challenges and opportunities in targeting and overcoming the endothelial barrier. The impediments in the clinical translation of vasculature organotropism in drug delivery are also discussed.

N-glycosylation by N-acetylglucosaminyltransferase IVa enhances the interaction of integrin beta 1 with vimentin and promotes hepatocellular carcinoma cell motility

Summary: N-glycosylation is closely associated with cancer metastasis. In this study, researchers found that GnT-IVa, a key transferase involved in forming N-glycans, promotes tumor growth and HCC cell motility by forming beta 1,4GlcNAc branches on ITGB1, a glycoprotein closely involved in cell motility by interacting with vimentin. The findings provide a novel mechanism for GnT-IVa in HCC progression and highlight the significance of N-glycosylation on ITGB1.

Hyaluronic acid: An overlooked extracellular vesicle contaminant

Summary: The variable presence of contaminants in extracellular vesicle (EV) samples is a major contributor to a lack of reproducibility in the field. Hyaluronic acid (HA) has been overlooked as a contaminant in EV fractions, but it is unexpectedly retained in some isolation methods. This should be considered in future studies to avoid potential inaccuracies.

Osteoarthritis and rheumatoid arthritis are the main diseases affecting joint cartilage, requiring long-term treatment for symptom relief. There is an urgent need for improved drug delivery strategies, particularly those based on micro- and nanotechnology. Encapsulation of therapeutic agents in delivery systems reduces drug efflux and protects against rapid clearance in the joint. The use of drug delivery systems can decrease side effects, increase therapeutic efficacy, and reduce the frequency of intra-articular administration.
