Inhibition studies with simple and complex (in)organic anions of the γ-carbonic anhydrase from Mammaliicoccus (Staphylococcus) sciuri, MscCAγ

Summary: The gamma-carbonic anhydrase (CA, EC 4.2.1.1) from Mammaliicoccus (Staphylococcus) sciuri (MscCA gamma) was studied for its inhibition by various (in)organic anions. Peroxydisulfate and trithiocarbonate were the most effective inhibitors, while other anions showed weaker inhibitory effects. Finding more effective MscCA gamma inhibitors could help combat antibiotic resistance.

Organoboronic acids/esters as effective drug and prodrug candidates in cancer treatments: challenge and hope

Summary: Boronic acids/esters have gained significant attention in medicinal and pharmaceutical research due to their unique structure, low toxicity, and ability to inhibit enzymes, capture cancer cells, and mimic certain types of antibodies. This review focuses on the potential of boronic acid/ester derivatives as pharmaceutical agents for treating multiple myeloma, prostate cancer, breast cancer, lung cancer, cervical cancer, and colon cancer. Although some newly developed boron-containing compounds have shown promising activities, further investigation is necessary before definitive conclusions can be drawn.

6-Substituted Triazolyl Benzoxaboroles as Selective Carbonic Anhydrase Inhibitors: In Silico Design, Synthesis, and X-ray Crystallography

Summary: In this study, substituted 6-(1H-1,2,3-triazol-1-yl)benzoxaboroles were synthesized and characterized using in silico design. The 6-azidobenzoxaborole was introduced as a molecular platform for preparing libraries of inhibitors through a click chemistry strategy. Compound 20 showed high selectivity as hCAVII and IX inhibitors with inhibition constants below 30 nM. Crystallographic investigation validated the design hypothesis and provided explanations for the different inhibition behavior observed against five hCA isoforms. Overall, compound 20 was identified as a promising lead compound to develop novel anticancer agents targeting hCA IX and potent neuropathic pain relievers targeting hCA VII.

Boron neutron capture therapy (BNCT) has emerged as a promising approach for cancer treatment with unprecedented precision. However, the clinical application of BNCT technology faces challenges due to the limitations of current drugs. Therefore, there is an urgent need to develop efficient boron agents with higher boron content and specific tumor-targeting properties.

The inhibitory effect of boric acid on hypoxia-regulated tumour-associated carbonic anhydrase IX

Summary: Carbonic anhydrases (CAs) are enzymes that catalyze the reversible hydration of CO2. One isoform, CA IX, is significantly overexpressed in various malignancies and is considered a validated target for new anticancer drugs. Inhibition of CA IX has been shown to decrease cancer cell proliferation and metastasis. Boric acid (BA) was tested as a CA IX inhibitor and was found to decrease proliferation of cancer cells in vitro, although the correlation between in vitro inhibition and in vivo effects was not strong. The mechanism of action of BA as an antiproliferative agent needs further investigation.

Cloning, purification, kinetic and anion inhibition studies of a recombinant β-carbonic anhydrase from the Atlantic salmon parasite platyhelminth Gyrodactylus salaris

Summary: A new beta-class carbonic anhydrase, named GsaCA beta, was cloned from the genome of the Monogenean platyhelminth Gyrodactylus salaris, a parasite of Atlantic salmon. This enzyme showed significant catalytic activity for the physiological reaction CO2 + H2O and was inhibited by various compounds. Since there are few non-toxic agents effective against this parasite, GsaCA beta is proposed as a new drug target.

Summary: Capillary electrophoresis was used to determine the thermodynamic acidity constants (pK(a)) and ionic mobilities of two types of carboranes, showing that the sulfamidoalkyl and sulfonamidoalkyl groups have weak acidity in the range of 10.78-11.45. The study also found that the ionic mobilities and hydrodynamic radii of the carborane anions varied depending on their valence.
