Congenital aniridia beyond black eyes: From phenotype and novel genetic mechanisms to innovative therapeutic approaches

Summary: Congenital PAX6-aniridia is not only characterized by the absence of the iris, but it is also associated with other ocular abnormalities and systemic features, making it a complex syndromic disorder. Foveal hypoplasia is now recognized as a more frequent feature than iris hypoplasia and plays a major role in visual prognosis. Genetic diagnosis of aniridia is challenging due to the genetic complexity caused by PAX6-related developmental genes. Targeted Next-Generation Sequencing combined with Whole-Genome Sequencing can increase the genetic diagnostic yield of aniridia.

Differential effects of experimental glaucoma on intrinsically photosensitive retinal ganglion cells in mice

Summary: This study investigates the survival of specific types of retinal ganglion cells (ipRGCs) and their associated functional deficits in glaucoma. The findings suggest that different subtypes of ipRGCs are differently affected by chronic ocular hypertension, with M4 ipRGCs involved in pattern vision being susceptible to the disease. This leads to reduced contrast sensitivity and visual acuity in mice with chronic ocular hypertension. On the other hand, M1 ipRGCs responsible for regulating circadian rhythmicity show minimal cell loss and therefore do not disrupt circadian re-entrainment in glaucomatous mice.
