Generation of reproducible model freshwater particulate matter analogues to study the interaction with particulate contaminants

Helene Walch et al.

Summary: Aquatic fate models and risk assessment require experimental information on the potential of contaminants to interact with riverine suspended particulate matter (SPM). However, the assumption of chemical equilibrium is invalid for particulate contaminants, and their interactions with SPM are governed by physicochemical forces between contaminant-particle and SPM surfaces. Therefore, the availability of a standard SPM material is highly relevant for the development of reproducible test systems to evaluate the fate of particulate contaminants in aquatic systems.

Determination of metallic nanoparticles in biological samples by single particle ICP-MS: a systematic review from sample collection to analysis

Adam Laycock et al.

Summary: A systematic review has found that current research on the use of single particle ICP-MS for analyzing engineered nanomaterials in biological samples is limited, with focus on a few types of materials and a lack of information for important tissues. Sample storage and appropriate protocol selection are critical for accurate analysis, and a one-size-fits-all approach is not practical. Efforts are needed to develop standardized methods and address issues such as nomenclature and control samples for regulatory use.

Determining nanoform similarity via assessment of surface reactivity by abiotic and in vitro assays

Didem Ag Seleci et al.

Summary: This work discusses the justification of grouping of nanoforms based on similarity of their surface reactivity, using four reactivity assays. A novel similarity assessment algorithm was applied to quantify similarities between pairs of nanoforms, with focus on the concentration-response in mass-metrics. Despite differences in outcomes from different reactivity assays, some pairs of case study nanoforms were considered sufficiently similar based on in vivo NOAECs, indicating conservative assessment by the abiotic assays.

Similarity assessment of metallic nanoparticles within a risk assessment framework: A case study on metallic nanoparticles and lettuce

Yuchao Song et al.

Summary: Similarity assessment is an effective method for using experimental data on nanoforms to regulate their fate and hazards. A case study found that chemical composition is the main driver of nanoform toxicity, while particle size had negligible impact on certain endpoints. Existing hazard data can be used to evaluate the hazards of similar nanoforms.

Assessing the similarity of nanoforms based on the biodegradation of organic surface treatment chemicals

Richard Cross et al.

Summary: This study presents a tiered testing strategy to assess the biodegradation of organic surface treatments used with nanomaterials. The approach allows for grouping of nanoforms with different organic surface modifications and reduces the burden of testing for environmental risk assessment of nanomaterials.

Reproducibility of methods required to identify and characterize nanoforms of substances

Richard K. Cross et al.

Summary: Nanoforms (NFs) can be distinguished by their physico-chemical properties. When registering NFs under the EU REACH framework, five basic descriptors are required for identification. Similarity assessment requires an understanding of achievable accuracy of the available methods. Reproducibility of six analytical techniques was assessed to estimate the achievable accuracy. Well-established methods performed well, while less mature technologies showed poorer reproducibility.

Integrated approaches to testing and assessment for grouping nanomaterials following dermal exposure

Luisana Di Cristo et al.

Summary: This study presents four grouping hypotheses for dermal toxicity of nanoforms (NFs) and develops tailored Integrated Approach to Testing and Assessment (IATA) for each hypothesis. The importance of considering NF dissolution in sweat fluids for dermal exposure is demonstrated. It is expected that these approaches will expedite the risk assessment of dermal toxicity for NFs.

Rationale and decision rules behind the ECETOC NanoApp to support registration of sets of similar nanoforms within REACH

Gemma Janer et al.

Summary: The new registration requirements for nanomaterials under REACH involve creating 'sets of similar nanoforms' for a more comprehensive human health and environmental assessment. A tool has been developed to assess the similarities between nanoforms based on intrinsic properties, with additional tiers for further evaluation if needed. The tool determines which nanoforms can be justified to be in the same set and provides a basis for REACH registration.

An integrated approach to testing and assessment of high aspect ratio nanomaterials and its application for grouping based on a common mesothelioma hazard

Fiona Murphy et al.

Summary: The development of an Integrated Approach to Testing and Assessment (IATA) aimed to support the grouping of different types of High Aspect Ratio Nanomaterials (HARNs) based on their potential to cause mesothelioma. The approach is hypothesis-driven, linking critical physicochemical descriptors for NFs to defined fate/toxicokinetic and hazard outcomes.

Creating sets of similar nanoforms with the ECETOC NanoApp: real-life case studies

Gemma Janer et al.

Summary: The ECETOC NanoApp is designed to assist industry in registering sets of similar nanoforms and to help regulators evaluate these registration dossiers. It systematically categorizes nanoforms based on their intrinsic properties and justifies their classification using functional properties. The decision rules are transparent and evidence-based, with a focus on human health hazards and risks.

The bioaccumulation testing strategy for manufactured nanomaterials: physico-chemical triggers and read across from earthworms in a meta-analysis

R. D. Handy et al.

Summary: Little is known about the bioaccumulation potential of manufactured nanomaterials, but a meta-analysis of existing data sets on particle properties and bioaccumulation in earthworms revealed that some particle metrics could trigger bioaccumulation testing and predict nano bioaccumulation factors. Additionally, nano biomagnification factors were calculated in rainbow trout tissue, showing significant correlations with earthworm tissue nano bioaccumulation factors. These findings suggest that bioaccumulation measurements in earthworms could be predictive of values in fish, allowing for confident prediction of the bioaccumulation potential of manufactured nanomaterials based on simple particle metrics.
