EMT-mediated regulation of CXCL1/5 for resistance to anti-EGFR therapy in colorectal cancer

Ye-Lim Park et al.

Summary: The emergence of RAS/RAF mutant clone is the main feature of EGFR inhibitor resistance in KRAS wild-type colon cancer. In this study, researchers successfully isolated cetuximab-resistant (CR) clonality cells from an in vitro system and found that these cells showed a higher EMT signature and increased expression of CXCL1/5. The study also revealed that CXCL1/5 expression in CR cells was regulated by SNAI2/NFKB and transactivated EGFR through CXCR/MMPI/EGF axis via autocrine signaling.

The Transcription Factor IRF9 Promotes Colorectal Cancer via Modulating the IL-6/STAT3 Signaling Axis

Bhesh Raj Sharma et al.

Summary: This study found that IRF9 plays an oncogenic role in CRC, and its loss reduces tumorigenesis. IRF9 promotes the activation of the pro-oncogenic STAT3 signaling pathway in the colon by promoting the release of IL-6. These findings provide new therapeutic strategies for the treatment of CRC.

The impact of VEGF on cancer metastasis and systemic disease

Yunlong Yang et al.

Summary: Metastasis, the leading cause of cancer-associated mortality, is still poorly understood. Both blood and lymphatic vasculatures play crucial roles in distal metastasis, with VEGF being a key angiogenic factor. Anti-VEGF drugs have been widely used in clinical treatment of various cancers, but their impact on cancer metastasis and systemic disease is largely unknown.

MDMX elevation by a novel Mdmx-p53 interaction inhibitor mitigates neuronal damage after ischemic stroke

Haomin Yan et al.

Summary: This study investigated the role of Mdmx in cerebral ischemia and the effects of a small-molecule Protein-Protein Interaction (PPI) inhibitor, K-181, on Mdmx-p53 interactions. The findings revealed that ischemic stroke decreased Mdmx expression and phosphorylation, while Mdmx overexpression showed a neuroprotective effect. The PPI inhibitor, K-181, attenuated neurological deficits by increasing Mdmx expression and selectively inhibiting HDAC6 activity.

Mutant erythropoietin enhances white matter repair via the JAK2/STAT3 and C/EBPβ pathway in middle-aged mice following cerebral ischemia and reperfusion

Rongliang Wang et al.

Summary: This study demonstrates that nonerythropoietic mutant EPO (MEPO) has neuroprotective effects on white matter injury in middle-aged male mice following cerebral ischemia by promoting microglial M2 polarization and oligodendrogenesis. This effect is achieved through the activation of JAK2/STAT3 and inhibition of the C/EBP beta signaling pathway.
