Thymol protects against bleomycin-induced pulmonary fibrosis via abrogation of oxidative stress, inflammation, and modulation of miR-29a/TGF-β and PI3K/Akt signaling in mice

Rasha M. Hussein et al.

Summary: This study found that thymol (THY) from thyme species can inhibit oxidative stress, inflammation, and lung fibrosis-related miR-29a/TGF-beta and PI3K/phospho-Akt signaling pathways. Repositioning itraconazole for amelioration of bleomycin-induced pulmonary fibrosis: Targeting HMGB1/TLR4 Axis, NLRP3 inflammasome/NF-κB signaling, and autophagy

Abeer Elkhoely et al.

Summary: This study investigated the potential mitigating effects of itraconazole on bleomycin-induced pulmonary fibrosis and explored the underlying mechanisms. The results showed that itraconazole had significant effects on oxidative stress, inflammation, high-mobility group box 1/toll-like receptor-4 Axis, autophagy, and nuclear factor kappa B/Nod-like receptor protein 3 inflammasome signaling. The neuroprotective and antidiabetic effects of trigonelline: A review of signaling pathways and molecular mechanisms

Youdan Liang et al.

Summary: The global epidemic of diabetes has led to increased pressure on public health, necessitating the development of new effective anti-diabetes strategies. Trigonelline, the main component of fenugreek, has been proven to have therapeutic effects on diabetes and its complications. The mechanisms by which trigonelline ameliorates diabetes include modulation of insulin secretion, reduction of oxidative stress, and improvement of glucose tolerance and insulin resistance. Additionally, trigonelline has been reported to possess neuroprotective properties against neurologic diseases such as Alzheimer's disease, Parkinson's disease, stroke, and depression. Comprehensive clinical trials are needed to evaluate the potential therapeutic effects of trigonelline.

Summary: Cellular senescence, especially the senescence of alveolar epithelial type II (ATII) cells, plays a crucial role in aging and aging-related diseases such as idiopathic pulmonary fibrosis (IPF). Plasminogen activator inhibitor 1 (PAI-1), a serine protease inhibitor, was found to bind to proteasome components and inhibit proteasome activity, leading to ATII cell senescence by preventing p53 degradation. This study provides insights into the molecular mechanisms underlying ATII cell senescence and its potential implications for the pathogenesis of lung fibrosis and other age-related diseases.

MicroRNA-27a-3p inhibits lung and skin fibrosis of systemic sclerosis by negatively regulating SPP1

Qi Cheng et al.

Summary: This study investigated the role and mechanism of miR-27a-3p in the fibrotic processes involved in systemic sclerosis (SSc). The results showed that miR-27a-3p is reduced in SSc lung and skin tissues. Overexpression of miR-27a-3p inhibited fibrosis-related genes expression and protein abundance, as well as cell proliferation. Furthermore, this effect was mediated by the negative regulation of SPP1 and ERK signal. These findings suggest that miR-27a-3p may be a potential target for delaying the progression of SSc fibrosis.

Summary: Fibrosing interstitial lung diseases are chronic and fatal lung diseases characterized by the irreversible accumulation of scar tissue in the lung tissue. Cellular senescence, defined as a cell fate decision caused by unrepairable cellular damage, plays an important role in the initiation and progression of pulmonary fibrosis, likely by promoting molecular and cellular changes in chronic fibrosing processes. Targeting cellular senescence may be a potential therapeutic approach for treating fibrosing ILDs.
